Abstract


Purpose by in vitro sperm adding different concentrations of Astragalus , Salvia , progesterone injection to study sperm calcium concentration . 28 specimens collected from outpatients were randomly divided into four groups : blank group , Astragalus group , Salvia group , progesterone group , each group added the same amount of different concentrations of reagents , Fluo3-AM passive diffusion into cells , the ester enzyme water to explain to release a Fluo3, Fluo3 calcium ion complexation , generated by the wavelength of 488 nm laser excitation and fluorescence , fluorescence values ??were positively correlated with Ca2 , determined by flow cytometry (FCM) and laser confocal (LSCM) sperm within Ca2 + concentration . All data analysis in SPSS 11.0 statistical software . Progesterone injection of 1.FCM detect the fluorescence intensity of Astragalus injection group sperm Ca2 and Salvia Injection , blank group difference was statistically significant ( P lt; 0.05 ) shows that progesterone injection vitro add Astragalus injection liquid allows the sperm Ca2 concentration increased ; 2 . Astragalus injection group , the fluorescence intensity was significantly enhanced in concentration ; Astragalus injection the group low concentrations significantly weaker fluorescence intensity , and high concentration comparison , the difference was statistically significant (P lt; 0.05); the progesterone injection group , in low concentration fluorescent group difference was not statistically significant (P gt; 0.05). 3.LSCM Determination of Astragalus injection group , progesterone injection group and Danshen injection group difference was statistically significant ( P lt; 0.05 ) ; Danshen injection group and blank group difference was not statistically significance ( P gt ; 0.05 ). Conclusion Astragalus injection can promote the sperm Ca2 influx and intracellular calcium concentration increases , the high concentration, in concentration the astragalus can significantly increase the sperm calcium level ; progesterone can promote the sperm Ca2 influx and intracellular calcium concentration increases , and luteal ketone drug concentration , the higher the concentration of the drug promote Ca2 influx capacity , the stronger ; Salvia sperm intracellular Ca2 concentration change is not obvious . That part of traditional Chinese medicine may increase sperm calcium concentration , thereby enhancing the in vitro motility of the sperm , artificial assisted reproductive chances of pregnancy .
