Abstract


Objective: Nephron sparing surgery of renal cell carcinoma (NNS) has been carried out extensively at home and abroad . Multi-center lesions is one of the important factors causing Nephron sparing surgery postoperative local recurrence , there are still certain recurrence rate , due . In a preliminary study of the experimental group has proved clear cell renal cell carcinoma (clear cell carcinoma, cc-RCC) is a monoclonal tumors , non- random X - chromosome in female patients , the most accurate marker for detecting tumor clonal origin inactivation (X-chromosome nonrandom inactivity). This study was designed by X-chromosome inactivation patterns , multi-center disease study renal clear cell carcinoma clone from . Methods: 11 female patients with renal cell carcinoma and corresponding normal kidney tissue specimens , 44 specimens were extracted DNA by methylation-sensitive restriction endonuclease enzyme Hha Ⅰ digestion of the human androgen receptor ( HUMARA) gene fragments by polymerase chain reaction (Polymerase Chain Reaction, PCR) , and gel electrophoresis , followed by a silver nitrate staining observed electrophoretic bands judgment cloned state . Results : HUMARA gene heterozygosity was 90.9% ( 10 / 11 ) . Clear cell renal cell carcinoma , the original living the same type , ie monoclonal origin was 90% ( 9 / 10 ) ; inactivation patterns between different polyclonal origin accounted for 10 % loss between the foci and multi-center foci (1 /10). Conclusion: The renal clear cell carcinoma lesions in the majority of the multi-center has the same X chromosome with the primary tumor inactivation patterns , suggesting that the lesions of the multi-center living type intrarenal transfer of the primary tumor , a small number of primary tumor of the same X chromosome loss suggestive of polyclonal origin , namely the primary tumor and the multi-center lesions occur independently . The above conclusion requires further large sample studies confirmed .
