Xuebijing endotoxin induced acute lung injury in rats with inflammatory response

Abstract


Objective: To investigate Xuebijing endotoxin (lipopolysaccharide, LPS)-induced acute lung injury (acute lung injury, ALI) influence the pathogenesis. Methods: 40 healthy male Wistar rats were randomly divided into four groups: normal control (NC) group :: 4ml/kg via the tail vein injection of saline (5 min inside); sham operation (S0) Group: along the midline to do a length of 1.5cm incision, remove the cecum, 2min after the cecum also satisfied the abdominal cavity, abdominal incision sutured and 4ml/kg tail vein injection of saline (5 min inside); cecal ligation and puncture (cecal ligation and puncture, CLP) Group: copy sepsis by cecal ligation model, and saline 4ml/kg tail vein injection (5 min inside); CLP Xuebijing: surgical method with the CLP group, postoperative Xuebijing 4ml/kg tail intravenous Note (5 min inside) in each group after molding 15 h, Arterial blood gas analysis rat arterial oxygen pressure (pressure of oxygen in arterial blood, PaO2); separation of serum by enzyme-linked immunosorbent assay (enzyme linked immunosorbent assay, ELISA) assay of serum tumor necrosis factor (tumor necrosis factor-α, TNF-α), cytokine interleukin-6 (interleukin-6, IL-6) and cytokine interleukin-8 (interleukin- 8, IL-8) of the content changes; observe the lung tissue hematoxylin and eosin (hematoxylin eosin, HE) staining results and conducting pairwise comparisons. Results: 1. PaO2 levels in rats compared: CLP PaO2 rats were significantly lower than NC group, S0 Xuebijing group and CLP group, the difference was statistically significant (P lt; 0.01), CLP Xuebijing lower than the NC group and S0 group (P lt; 0.01). (2) serum levels of IL-6 compared: CLP serum levels of IL-6 was significantly higher than NC group, S0 Xuebijing group and CLP group, the difference was statistically significant (P lt; 0.01), CLP Xuebijing group than NC group and S0 group (P lt; 0.01). 3 serum levels of IL-8 Comparison: CLP serum levels of IL-8 was significantly higher than NC group, S0 Xuebijing group and CLP group, the difference was statistically significant (P lt; 0.01), CLP Xuebijing group than NC group and S0 group (P lt; 0.01). 4 of serum TNF-α levels in comparison: CLP of serum TNF-α were significantly higher than NC group, S0 Xuebijing group and CLP group, the difference was statistically significant (P lt; 0.01), CLP Xuebijing group than NC group and S0 group (P lt; 0.01). 5 HE staining of lung tissue comparison: CLP rats observed widened alveolar septum, pulmonary interstitial see large amounts of inflammatory cell infiltration, alveolar lumen narrowing, bleeding, exudation of neutrophils and inflammatory exudate. CLP Xuebijing exudation of neutrophils and inflammatory exudate was reduced. Conclusion Xuebijing can reduce endotoxemia-induced lung inflammation, endotoxin-induced acute lung injury (ALI) has a protective effect when the lungs.
