Abstract


Objective:Fluid Shear Stress (FSS) may lead to positive bone remodeling through effects on both osteoblasts and osteoclasts. In this study, the effects of FSS on expression of osteoprotegerin (OPG) and osteoclast differentiation factor(ODF) that is receptor activator of NF-kB ligand (RANKL), two important regulators of bone remodeling, were investigated.Methods:We developed an in vitro model to load MC3T3-E1 and studied levels of OPG and RANKL. Cells were exposed to various durations of FSS, then cells was lysated and levels of OPG and RANKL were quantified using Immunofluorescence and Western Blotting.Results:We found that FSS signitificantly increased OPG levels. Levels of RANKL detected were not signitificantly affected by FSS. OPG:RANKL ratio was signitificantly increased.Conclusions:The relative shift in abundance OPG over RANKL associated with applied FSS stimulation suggest the OPG:RANKL ratio may be important regulators of load-induced bone formation/resorption by coupling osteoblastic and osteoclastic mechanisms.
