Abstract


Objective To investigate the multiple intrathecal injection of different doses of nitric oxide (nitric oxide, NO) precursor L-arginine (L-Arg) and nitric oxide synthase (nitric oxide synthase, NOS) inhibitor Nω-nitro-L - arginine methyl ester (Nω-nitro-L-arginine methylester, L-NAME) inflammatory behavior in the rat formalin pain reaction Bcl-2/Bax expression in the dorsal horn of the spinal cord. bcl-2 gene to inhibit apoptosis, bax promote apoptosis gene, both the ratio of the amount of expression can be used as a judgment indicator of apoptosis. The analysis of the application of different doses for a long time relationship between NO and inflammatory pain apoptosis, to explore the role of NO in pain modulation and its mechanism. 24 healthy about 120 g SD rats were randomly divided equally into four groups, each group of animals were intrathecal injection of normal saline (NS), different doses of NO precursor L-arginine 10μg / d (low-L- Arg group) and 250μg / d (high L-Arg group), and the nitric oxide synthase inhibitor Z-NAME2700μg / d (L-NAME group). Each group of animals each administered once a day for four consecutive days. All animals 20 minutes after administration of the four days, in rat right hind limb feet subcutaneous injection of formalin (2%, 0.1 ml), followed by one hour rats licking licking and biting behavior of pain reaction. Four hours after decapitation lumbar enlargement of the spinal cord L4-5 segment, Immunohistochemical detection of spinal dorsal horn of c-Fos protein and neuron nitric oxide synthase (nNOS) expression of Bcl-2 and Bax protein the number of positive cells; expression using Western-blot detection of spinal cord Bcl-2 and Bax protein content. Results (1) formalin rat licking licking and biting behavior in experimental pain response two phases. The results show that in each group of the first phase of acute pain behavioral responses no significant differences; each group phase chronic pain behavioral responses accumulated time low L-Arg group decreased significantly compared with the NS group, L-Arg group and L -NAME group than in the NS group increased significantly. (2) immunohistochemistry results showed that: 1) spinal cord dorsal horn superficial Fos protein and nNOS positive cells compared with NS both low L-Arg group were reduced, while the L-Arg group both increased L-NAME group Fos expression increased and decreased nNOS expression; 2) spinal cord dorsal horn shallow of Bcl-2 and Bax positive cell count, low L-Arg group both increased compared with NS group, high L-Arg of Bcl-2 reduced Bax increased, the average number of L-NAME group both. (3) The immunohistochemistry results showed that Bcl-2 and Bax, the ratio of the number of positive cells, and Western-blot results showed that the ratio of Bcl-2 and Bax protein content in each experimental group and the control group compared with the low L-Arg group Bcl-2/Bax ratio was significantly higher Bcl-2/Bax ratio of L-Arg and L-NAME group were significantly reduced. Conclusions (1) several different doses of NO has a double-sided, small doses of spinal cord pain modulation has analgesic effect of large doses has the role of promoting pain, lack of NO production also has the role of promoting pain; (2) a small dose of NO inhibit apoptosis, and large doses of NO can promote apoptosis, while insufficient NO production can also cause apoptosis; (3) speculated that a small dose of NO may inhibit apoptosis to produce analgesia, large doses of NO may promote apoptosis, resulting in the role of promoting pain, lack of NO production may promote apoptosis promote pain resulting. The definitive evidence needs to be further to explore.
