Objective: To compare chitooligosaccharides on human liver cells (Chang Liver) and human hepatoma cells (SMMC-7721) the impact of the induction of apoptosis, and to explore the molecular mechanisms, in order for the clinical development of new drugs of low toxicity and side effects of anti-hepatoma provide experimental basis. Methods: MTT assay chitooligosaccharides different concentrations and different time, liver cells and hepatoma cell proliferation inhibition rate; Hoechst 33258 fluorescent staining chitooligosaccharides role, the morphological changes of the liver cells and liver cancer cells; flow cytometry After the instrument detection Oligochitosan role, hepatocytes and hepatoma cell apoptosis rate; by Western blotting detect chitooligosaccharides role, liver cells and liver cancer cells to apoptosis related protein Bcl-2.Caspase-3 expression changes. Results: 1) 0.5-2.5 mg / ml chitooligosaccharides respectively role for 24 h and 48 h after the liver cancer cell proliferation by a different degree of inhibition, and there is a dose-dependent effect, the 50% inhibitory concentration (50% concentration of inhibition, with an IC50 ) of 2.5 mg / ml, and the proliferation of liver cells were not suppressed; 3.0-4.0 mg / ml chitooligosaccharides role for 24 h, 48 h, the proliferation of liver cells and liver cancer cells was inhibited, but hepatoma cell inhibition rate was significantly greater than the hepatocytes. 2) 2.5 mg / ml chitooligosaccharides role after 24 h, liver cells marked apoptosis occurred, no significant changes in the liver cells. 3) 2.5 mg / ml of chitooligosaccharides role after 24 h, the rate of apoptosis of hepatoma cells was significantly increased to 76.04%, no significant change in the rate of liver cell apoptosis. 4) 2.5 mg / ml chitooligosaccharides role after 24 h, Bcl-2 expression in hepatoma cells significantly reduced Caspase-3 expression was significantly increased; while no significant change in the expression levels of the two proteins in liver cells . Conclusion: chitooligosaccharides on human liver cells (Chang Liver) and inhibit the proliferation of human hepatoma cells (SMMC-7721) exist significant differences, low concentrations of chitooligosaccharides significant inhibition of proliferation of liver cancer cells, liver cells The proliferation was no significant inhibition. Chitooligosaccharides there are obvious differences induced apoptosis of liver cells and liver cancer cells, low concentrations of chitooligosaccharides significant pro-apoptotic effect on liver cells, but can significantly promote apoptosis of hepatoma cells and down suppression withered apoptotic protein Bcl-2 and the upward promote apoptotic protein Caspase-3 expression. The chitooligosaccharides induced apoptosis of liver cells and liver cancer cells showed a cell selective, which may be related to the interaction of cells and chitooligosaccharides and structure. Chitooligosaccharides on human liver cells (Chang Liver) and the induction of apoptosis of human hepatoma cells (SMMC-7721) were compared, and to explore the molecular mechanism, low toxicity and efficient for the clinical development of anti-liver cancer drug an experimental basis and reference.
