Oral squamous cell carcinoma (OSCC) is one of the common malignant tumors of the oral and maxillofacial region, with a high recurrence and high transfer rate. Cancer stem cell theory in recent years become a hot topic in vitro cell culture studies have shown that CD44, podoplanin and ABCG2 and as candidate biomarkers of cancer stem cells, and squamous cell growth, metastasis and recurrence with a high degree of correlation, but in oral squamous cell tumors positioning and distribution have yet to see the study. Objective: To establish OSCC of new treatment strategies, we detect the cancer stem cell candidate signs matter of CD44 Expression of podoplanin and ABCG2 in OSCC in the expression of status, analyze its with OSCC occurrence, histological type and clinical prognosis of correlation: In this study, using immunohistochemical staining method for detection of CD44 in 30 cases of human cases of OSCC tissues, podoplanin and ABCG2 protein expression, to observe distribution and positioning. Experimental data using SPSS11.5 software for statistical analysis of the results: the expression and localization of the 30 patients with oral squamous cell carcinoma, CD44-positive expression in 28 cases, the positive expression rate was 93.33%; Expression of podoplanin positive expression in 21 cases, the positive expression rate of 70 %; ABCG2 positive expression of the 24 cases, the positive rate of 80%. (1) adjacent epithelial cancer, CD44 expression in the cytoplasm and cell membrane of the basal cell layer and the spinous layer, podoplanin expression in the cytoplasm and cell membrane of the lymphatic vessels near the basal cell layer, and ABCG2 only in epithelial keratosis layer The specific area visible expression of a strong film. (2) in well-differentiated cancer nests, CD44 and podoplanin expression patterns similar to the expression patterns in adjacent epithelial membrane staining was very strong in the center of the cancer nest, ABCG2 expression or weak expression in the tumor area, CD44 still was strong membrane staining in the tumor stromal lymphocytes. Poorly differentiated cancer nests, CD44, podoplanin and ABCG2 are positive expression. (3) In cases of chemotherapy, CD44, podoplanin and ABCG2 expression around the necrotic cancer nests. Histological type CD44 and ABCG2 expression was correlated with the degree of differentiation is no significant correlation (P gt; 0.05); Expression of podoplanin positive expression with differentiation decreased significantly higher (P lt; 0.05) and clinical prognosis correlation (1) of CD44 of ABCG2 and podoplanin expression in oral squamous cell carcinoma of gender, age, T stage, the effect of chemotherapy, and clinical stage there is no relationship (P gt; 0.05) (2) of ABCG2 expression in lymphoid its expression rate of metastasis and without metastasis group were 93.75% and 64.29%, respectively, both the presence of significant differences (P lt; 0.05) (3) Expression of podoplanin expression in lymphatic metastasis group and no forwarding group expression rates were 87.5% and 50%, both the presence of a significant difference (P LT; 0.05). Conclusion: 1. Cancer stem cell marker substance ABCG2 and podoplanin and lymphatic metastasis has a significant correlation between CD44 and podoplanin in tumor tissue after chemotherapy expression overlap, suggesting that tumor stem cell markers can be used as the observations of clinical outcomes. 2. Podoplanin with the degree of tumor differentiation and relevance, and the cancer stem cell marker CD44, podoplanin and ABCG2 expression in poorly differentiated carcinoma high degree of overlap, suggesting that the cancer stem cell markers can be used as the observations of the degree of differentiation.
