Abstract


Objective: To study the sporadic colorectal cancer of hMLH1 of hMSH2 expression and COX-2 expression, and to investigate the expression and biological characteristics of colorectal cancer. Methods: Using immunohistochemistry method (PV-9000 two-step method) detected 52 cases of sporadic colorectal cancer, 22 cases of colorectal adenomas, 20 cases of normal mucosa of hMLH1, hMSH2, COX-2 expression. According to the dyeing depth and the number of positive cells score and calculated ratings ,0-2 is divided into the positive (negative (-), 3 more points). To apply x2 test analysis of hMLH1, hMSH2, COX-2 in sporadic colorectal cancer, colorectal adenomas, the expression in the normal mucosa and the relationship with the biological characteristics of sporadic colorectal cancer; application pearson correlation analysis of hMLH1, hMSH2 and COX -2 expression in sporadic colorectal cancer. Results: 1.hMLH1 of staining was mainly localized in the nucleus. HMLH1 expression in sporadic colorectal cancer reduced. In normal mucosa, colorectal adenomas, sporadic colorectal carcinoma rate was 100.00%, 77.27%, 75.00%, and was gradually decreased. Sporadic colorectal cancer, colorectal adenomas and normal mucosa group, a significant difference (P <0.05). No significant differences between sporadic colorectal cancer and colorectal adenomas. hMSH2 was localized in the nucleus, reduced hMSH2 expression in sporadic colorectal cancer. In normal mucosa, colorectal adenomas, sporadic colorectal carcinoma rate was 100.00%, 82.82.0%, 78.85%, and was gradually decreased. Sporadic colorectal cancer, colorectal adenomas and normal mucosa group, a significant difference (P lt; 0.05). No significant differences between sporadic colorectal cancer and colorectal adenomas. 2.hMLH1 of hMSH2 expression with sporadic colorectal cancer biological characteristics: hMLH1, of hMSH2 expression of gender, age, tumor size, tumor location, lymph node metastasis, Dukes stage no correlation; related to the degree of tumor differentiation. hMLH1, hMSH2 in poorly differentiated carcinoma group, the positive expression rate of 55.56% and 61.11%, respectively, lower than the high - differentiated cancer group, the positive expression rate of 85.29% and 88.24%, the difference was statistically significant. 3.COX-2 positive expression mainly in the cytoplasm, in the normal mucosa, colorectal adenomas sporadic colorectal carcinoma increased gradually. COX-2 expression in sporadic colorectal cancer group was 78.85%, significantly higher than that of colorectal adenomas, 54.55% and 5.00% of the normal mucosa group. COX-2 in sporadic colorectal cancer, colorectal adenomas, normal mucosa group differences were statistically significant (P <0.05). Expression with sporadic colorectal cancer biological characteristics of 4.COX-2: COX-2 expression in sporadic colorectal cancer and sex, age, tumor size, tumor location and tumor differentiation degree of correlation with lymph node metastasis Dukes stage correlation. COX-2 expression in lymph node metastasis group rate of 89.28% in sporadic colorectal cancer tissues was significantly higher than the free expression of the lymph node metastasis rate of 66.67%, the difference was statistically significant. Dukes C, D phase of COX-2 expression rate of 92.86% was significantly higher than that in Dukes A, B of the expression rate of 62.50%, the difference was statistically significant. 5. HMLH1, hMSH2 and correlation of COX-2 expression in sporadic colorectal cancer: COX-2 positive expression of 41 specimens, hMLH1 positive expression in 36 cases, five cases with negative expression; expression of COX-2 negative, 11 cases specimens, hMLH1 positive expression in the three cases, the negative expression of eight cases. The correlation analysis, r = -0.571, P <0.01. 41 specimens positive expression of COX-2, hMSH2 positive expression of the 37 cases, 4 cases; negative expression of COX-2 negative expression in 11 specimens, hMSH2 positive expression of the four cases, seven cases of negative expression. Analysis by means of correlation r = -0.539, P <0.01. So of hMLH1, of hMSH2 expression of COX-2 expression was negatively correlated. Conclusion: 1.hMLH1, hMSH2 expression reduce the incidence of colorectal cancer, the development is closely related to, and its expression is associated with the degree of differentiation of the colorectal cancer. 2.COX-2 in the incidence of colorectal cancer process may play an important role in its expression and colorectal cancer lymph node metastasis, Dukes staging. 3 of sporadic colorectal cancers in hMLH1, hMSH2 expression of COX-2 expression was negatively correlated.
