Objective: To investigate cyclin E (of CyclinE), matrix metalloproteinase -9 (MMP-9), and 10 chromosome deletion phosphatase and tensin homologue (PTEN) gene expression of survivin (Survivin) the correlation of the clinical and pathological features of colorectal cancer, and want to be able to more accurately determine the prognosis of patients with colorectal cancer, and to provide a theoretical basis to find the corresponding gene therapy method. PATIENTS AND METHODS: The immunohistochemical Envision two-step method to detect 12 cases of colorectal normal mucosa and 66 cases of colorectal cancer tissue of CyclinE, MMP-9, Survivin and PTEN expression to study the expression of these four proteins and the large intestine cancer patients, gender, age, tumor location, macroscopic type, depth of invasion, the relationship between the degree of differentiation, Dukes stage and lymph node metastasis, and analyzed statistically. Results: 1,12 normal intestinal mucosa, CyclinE cases were positive, MMP-9 3 cases were positive, the Survivin positive expression, PTEN 11 cases of positive expression. 2,66 cases of colorectal cancer of CyclinE,, MMP-9, Survivin protein expression positive rate were 62.12%, 75.76%, 71.21%, significantly higher than the expression in normal colorectal tissue (8.33%, 25.0%, 0) the difference was significant (P lt; 0.01); positive expression of PTEN in colorectal cancer rate was 53.03%, significantly lower than the expression in normal colon mucosa (91.67%), the difference was significant (P lt; 0.05). 3 of CyclinE abnormal expression of colorectal cancer Dukes stage and lymph node metastasis related (P lt; 0.05); MMP-9 expression and depth of invasion, tissue differentiation, Dukes stage and lymph node metastasis were related (P lt; 0.05 ); Survivin expression and Dukes stage and lymph node metastasis was significantly related (P lt; 0.05); PTEN expression and depth of invasion of colorectal cancer, Dukes stage and lymph node metastasis was significantly related (P lt; 0.05). 4, correlation analysis showed that: the the colorectal in PTEN of expression and of CyclinE, MMP-9 in the expression of Survivin was significant with a negative correlation between, on PTEN and of CyclinE, MMP-9 in, of Survivin in colorectal cancer occurrence development from the antagonistic role; MMP-9 protein expression increased with the enhancement of Survivin protein expression intensity, both showed a significant positive correlation (r = 0.447). Research conclusions: 1 of CyclinE, expression of MMP-9 and Survivin in colorectal cancer expression significantly increased, of PTEN in colorectal cancer expression significantly with reducing and illustrate them in the incidence of colorectal cancer, development, infiltration and the transfer process from the important role . 2, CyclinE, MMP-9, Survivin and PTEN gene protein expression and clinicopathological features of relevance to their expression level can be used as the detection of colorectal useful indicator of the degree of malignancy. Of PTEN expression and of CyclinE colorectal MMP-9 in, the expression of Survivin was significantly with negative correlation, the PTEN gene expression down and of CyclinE, MMP-9 and Survivin gene expression raised possible in the incidence of colorectal cancer, the development process in the synergies. The expression of MMP-9 and Survivin expression was significantly positive correlation between them in a synergistic inhibition of apoptosis and promote tumor angiogenesis or mutual adjustment mechanism, and through various channels to promote the occurrence and development of colorectal cancer. 4, the joint detection of four genes protein expression levels of great value to assess the biological behavior of colorectal helpful in assessing the prognosis of patients, to provide the basis for clinical treatment, may become the site of colorectal cancer molecular targeted therapy.
