Objective To investigate the tumor suppressor gene RUNX3 protein expression in gastric cancer tissues, and clinical significance of the comparative analysis of the two, in order to understand the role of the tumor suppressor gene RUNX3 in gastrointestinal tumors. The collection of 52 surgically resected gastric cancer patients and the same period inpatient surgical specimens of 41 patients with rectal cancer ① HE staining of gastric cancer and colorectal cancer tissue and adjacent normal tissue morphological changes; ② SP immunohistochemical assay of tumor suppressor gene RUNX3 protein expression in gastric cancer tissues and clinical analysis; ③ computer image analysis Contact RUNX3 protein expression in gastric cancer tissues; ④ Western blot detection RUNX3 protein expression in gastric cancer tissues; ⑤ SPSS for Windows13.0 statistical software processing image analysis data immunohistochemical chemical average optical density values ??and Western blot gray value. Results ① HE staining of the stomach wall of the adjacent normal tissues: cancer, rectal wall four layers from the inside out, the mucosa, submucosa, muscularis, adventitia, mucosal layer is divided into the epithelium, lamina propria, muscularis mucosa. Gastric carcinoma HE staining, histological type adenocarcinoma, a common tubular adenocarcinoma; cancer histological type is more common in well-differentiated tubular adenocarcinoma and papillary adenocarcinoma. ② immunohistochemistry: next to the corresponding normal tissue in 52 cases of gastric RUNX3 protein could be expressed, surface mucous cells are mainly located in the cytoplasm of the gastric glands; 52 cases of gastric carcinoma in 32 cases (61.5%) of RUNX3 protein expression positive gastric cancer differentiation level is lower, the less positive expression, and the presence of cytoplasmic and nuclear staining heterogeneity; corresponding normal tissue adjacent 41 cases of rectal mucosa RUNX3 protein expression in 38 cases (92.7%), located in the columnar epithelial cells cytoplasm, nucleus, 41 patients with rectal carcinoma in 19 patients (46.3%) positive expression below the gastric the RUNX3 protein positive rate (P <0.05); immunohistochemical image analysis shows that the OD value of RUNX3 protein expression in gastric cancer group better than the cancer group, the difference was statistically significant (P <0.05). ③ Western blot results showed that 52 cases of gastric carcinoma the RUNX3 protein of 38 cases (73.1%) were down-regulated or absent, poorly differentiated gastric cancer, RUNX3 protein expression was significantly higher than that in normal gastric mucosa of the gastric tissue (P <0.05) ; 41 cases of rectal cancer RUNX3 protein expression was positive in 15 cases (36.6%), poorly differentiated cancer, RUNX3 protein expression in normal rectal mucosa was significantly higher than the corresponding cancer tissues (P <0.05). Conclusion ① HE staining showed that gastric cancer, rectal cancer, next to the corresponding normal tissues (away from cancer primary tumor more than 10cm) can be used as a normal control group; gastric adenocarcinoma tissue types, the common tubular adenocarcinoma; rectal cancer in high differentiated tubular adenocarcinoma and papillary adenocarcinoma is more common. ② gastric cancer tissues compared with normal gastric mucosa tissue RUNX3 protein expression rate was significantly lower, suggesting that RUNX3 protein downregulation may be related with gastric cancer development. ③ the RUNX3 protein expression in cancer tissues than paraneoplastic mucosal tissue was significantly reduced, suggesting that downregulation of RUNX3 protein may be associated with the occurrence of colorectal cancer related to the development. OD value of ④ RUNX3 protein expression in gastric cancer group than cancer group, suggesting that RUNX3 protein downturn is likely in the diagnosis of gastric cancer is more meaningful than rectal cancer. ⑤ RUNX3 protein expression in gastric cancer patients, gender, age, tumor location, depth of invasion, size, Lauren classification has nothing to do with the degree of differentiation and lymph node metastasis; of RUNX3 protein expression and rectal cancer patients, gender, age, tumor location, size has nothing to do with the degree of differentiation, tumor depth of invasion, Dukes stage and lymph node metastasis. (6) from the morphological and molecular level depth Runx3 structure and function of the gene expression regulation mechanisms and regulatory networks gastrointestinal tumorigenesis new biological markers and tumor gene therapy for the diagnosis and treatment of gastrointestinal tumors targets.
