Abstract


Purpose ① understand HBsAg-positive pregnant women in placental cells apoptosis. (2) explore the relationship between TLR3 and of placental apoptosis placenta HBV infection. ③ explore the role of of TLR3 placental apoptosis in HBV intrauterine infection. 136 cases. HBsAg-positive pregnant women at all levels by the Taiyuan hospital screening from July 2003 to June 2004, the prenatal examination and childbirth, obstetrics and gynecology at the Infectious Diseases Hospital of Taiyuan as the research object, select the same period in Shanxi Medical Clinical Hospital prenatal check-free pregnancy and childbirth infection, 30 cases of normal pregnant women without pregnancy complications as control. Collection of HBsAg-positive pregnant women and normal pregnant women placental tissue, collecting 2 ml of HBsAg-positive pregnant women in the venous blood of their newborns femoral vein and to collect epidemiological baseline study material at the same time. The following studies: (1) using ELISA detection of serum HBsAg, quantitative PCR Quantitative serum HBV DNA; ② by immunofluorescence double labeling was detected in placental tissue HBsAg and of TLR3; ③ using the Tunel assay placenta cells apoptosis . . Screening by Taiyuan levels Hospital from December 2008 to January 2009, obstetrics and gynecology at the Infectious Diseases Hospital of Taiyuan prenatal check and the childbirth detection of HBV intrauterine infection HBsAg-positive pregnant women in 20 cases, same period in childbirth match the proportion of 1:3 and 1:2, respectively, non-intrauterine infection in pregnant women, 60 cases and 40 cases of normal pregnant women. Placental tissue collected aseptically pregnant women, and the extraction of total RNA, 2 ml of acquisition of HBsAg-positive pregnant women in the venous blood of their newborns femoral vein. The following studies: (1) using ELISA detection of serum HBsAg quantitative PCR Quantitative serum HBV DNA; ② Real-time RT-PCR detection of TLR3 mRNA and caspase3 mRNA, expression. Results ① used the Tunel method to detect placental apoptosis group apoptotic index the: _HBsAg positive pregnant women apoptotic index was significantly higher than in normal pregnant women (t = 37.834, P lt; 0.001), but each serum HBV DNA levels were apoptotic index No statistically significant difference (F = -0.727, p = 0.487); placenta HBV infection apoptotic index was significantly lower than the placenta non-infected group (t = 2.614, p = 0.010); of TLR3 positive group TLR3 negative apoptotic index no significant difference (t = 1.162, p = 0.247); apoptotic index of HBV intrauterine infection group and non-intrauterine infection group was no significant difference (t = -1.956, p = 0.053). (2) using real-time quantitative RT-PCR, full-term placental of TLR3 mRNA caspase3 mRNA expression compared group differences. TLR3 mRNA relative expression level: HBsAg-positive pregnant women group than normal pregnant group, the difference was statistically significant (t = -21.714, P lt; 0.0001), but no significant difference in of each serum HBV DNA levels were (F = 0.727, P = 0.487); HBV intrauterine infection group and non-intrauterine infection group was no significant difference (t = 1.685, P = 0.096). relative caspase3 mRNA expression levels: HBsAg-positive pregnant women group than normal pregnant group, the difference was statistically significant (t = -2.467, P = 0.015), but each serum HBV DNA levels were no statistically significant difference (F = 1.151, P = 0.322); HBV intrauterine infection group and non-intrauterine infection group was no significant difference (t = 1.826, P = 0.072). ③ term placenta TLR3 mRNA and caspase3 mRNA, relative expression levels of correlation analysis showed that: normal pregnant group, Caspas3 Ct / GAPDH Ct value, and TLR3_Ct/β-actinCt value was negative correlation (r = -0.479, p = 0.002); HBsAg-positive pregnant women in group two variables found no correlation (r = 0.106, p = 0.351), the correlation between the two variables further intrauterine infection group and non-intrauterine infection group were There was no statistically significant (r = -0.152, p = 0.512; r = 0.162, p = 0.221). Conclusion ① normal pregnant women at term of placental apoptosis index and relative caspase3 mRNA expression were lower than HBsAg-positive pregnant women, HBV can cause increased placental apoptosis. (2) HBsAg positive pregnant women, the placenta HBV infection apoptotic index was significantly lower than the placenta of non-infected group, suggesting that placental apoptosis may be protective factors of HBV placental infection. (3) normal pregnant women at term placenta TLR3 mRNA relative expression level significantly lower than serum HBsAg positive pregnant women, suggesting that HBV infection can up-regulate the expression of placental TLR3. ④ serum HBsAg positive pregnant women did not find that there is a correlation of TLR3 and placental apoptosis.
