Objective: To establish the experimental type 2 diabetic rat model and applied to psychological stress, by comparing the different groups of rats Thl type cytokines (IL-2) and Th2 cytokines (IL-4) and IL-18 in serum in the content, to explore the impact of psychological stress on the immune function of diabetic rats and its mechanisms. Methods: an experimental type 2 diabetic rat model. 60 rats adapt to a week later, the rats were randomly divided into the model (40) and health (20), the model with high-fat diet plus low-dose intraperitoneal injection of streptozotocin multiple times streptozotocin (STZ) to establish the experimental type 2 diabetic rat model. 2, the implementation of psychological stress. Type 2 diabetic rats criteria to reach blood sugar random principle to extract the 20 model group, and were randomly divided into model control group and model stress group, healthy rats were randomly divided into the healthy control group and health stress group . Forced cold swim and restraint stress model stress group and health stress group psychological stress. The results: 1. Experimental type 2 diabetic rat model. Made the module injection of STZ Two days later, a random sample of model rats fasting glucose to 18.739 ± 2.160 (mmol / l) to achieve the criteria of experimental rat model of type 2 diabetes. By intraperitoneal blood after the end of the experiment, the model group and the healthy control group rat insulin (IU / ml) were 26.294 ± 5.928 (IU / ml) and 33.294 ± 2.382, P = 0.020; glucagon 380.607 ± 64.128 (pg / ml) and 296.308 ± 68.759 (pg / ml), P = 0.036; insulin sensitivity index were -6.021 ± 0.345 and -5.044 ± 0.158, P = 0.000. In line with the characteristics of the experimental type 2 diabetic rats indicators of glucose metabolism. 2. Rats IL-2 test results. Health stress group and the healthy control group rat IL-2 levels were 159.504 ± 65.464 (ng / ml) and 111.124 ± 33.348 (ng / ml), the model stress group and model control group rat IL-2 levels were 176.848 ± 61.462 (ng/m1) and 221.015 ±. 48.847 (ng/m1) A factor main effects P = 0.017, the factors B main effect, P = 0.531. rats IL-4 test results. Health stress group and the healthy control group rat IL-4 levels were 42.394 ± 9.923 (pg / ml) and 36.781 ± 6.035 (pg / ml), the model stress group and model control group rat IL-4 levels were 64.528 ± 8.388 (pg / ml) and 53.317 ± 10.834 (pg / ml), the factors A main effect P = 0.023, the the factors B main effect P = 0.735; each group rat IL-18 test results. The health stress group and health control rats, the level of IL-18 were 65.514 ± 15.362 (ng/m1) and 57.618 ± 13.607 (ng / ml), the model stress group and model control group IL-18 levels were 96.652 ± 15.501 (ng / ml) and 77.147 ± 12.235 (ng / ml), the factors A main effect P = 0.007, the factors B main effect, P = 0.207; 5. the the model stress group and model control group, IL-2, IL- 4 and IL-18 in comparison. The model stress group and model control group rat IL-2 levels were 176.848 ± 61.462 (ng / ml) and 221.015 ± 48.847 (ng / ml), P = 0.195; IL-4 levels were 64.528 ± 8.388 (pg / ml) and 53.317 ± 10.834 (pg / ml), P = 0.026; IL-18 levels were 96.652 ± 15.501 (ng / ml) and 77.147 ± 12.235 (ng / ml), P = 0.030. Conclusion: 1. Experimental type 2 diabetic rats preparation process can lead to changes in immune function in rats. Psychological stress on the immune function of healthy rats. 3 significant effect of psychological stress on experimental type 2 diabetic rat model Thl type cytokines (IL-2), but have a significant impact on Th2-type cytokines (IL-4). Psychological stress can change the level of IL-18 in experimental type 2 diabetic rats.
