Abstract


Objective: To detect different tidal volume mechanical ventilation in rat lung tissue high mobility group protein 1 (HMGB1) mRNA and protein expression level and Xuebijing injection on tidal volume mechanical ventilation rat lung tissue HMGB1 explore HMGB1 ventilator-induced lung injury (VILI) incidence in role, and Xuebijing the injection intervention VILI role. Methods: In this study, the experiment is divided into two parts: 24 healthy male Wistar rats were randomly divided into three groups, namely the control group, low tidal volume (L-VT) group and tidal volume (H-VT) group. Myeloperoxidase assay kit each rat plasma and bronchoalveolar lavage fluid (BALF) and myeloperoxidase (MPO) activity, using immunohistochemical method (SABC method) and in situ hybridization detection lung tissue HMGB1 expression levels. Experiment II: 24 healthy male Wistar rats were randomly divided into three groups, the control group, tidal volume group, the blood will be net intervention group. Myeloperoxidase assay kit MPO activity in rats plasma and bronchoalveolar lavage fluid, using immunohistochemical method (SABC method) and in situ hybridization detection of lung tissue HMGB1 expression levels. Results: Experiment 1: 1, tidal volume group rat plasma and BALF MPO activity was significantly higher than that in the control group and the small tidal volume group (P lt; 0.01), control group with low tidal volume group difference was not statistically significant (P GT ; 0.05). 2, tidal volume lung tissue airway epithelial cells HMGB1 mRNA and protein expression level was significantly higher and low tidal volume group (P lt; 0.01), low tidal volume group and the control group, the difference was not statistically significant (P gt; 0.05). Experiment II: 1, tidal volume group and the blood will be a net intervention group rat plasma and BALF MPO activity was significantly higher (P lt; 0.01); tidal volume group rat plasma and BALF MPO activity is higher than the blood will be net intervention group (P lt; 0.01). 2, tidal volume group and the blood will be a net intervention of the lung tissue airway epithelial cells HMGB1 mRNA and protein expression level was significantly higher (P lt; 0.01); tidal volume of the lung tissue airway epithelial cells HMGB1 mRNA and protein expression level higher than Xuebijing to the intervention group (P lt; 0.01) Conclusion: 1, tidal volume ventilation-induced lung tissue HMGB1 mRNA and its protein highly expressed HMGB1 secretion led to an increase in the inflammatory response of the lung tissue expansion, may be ventilator-induced lung injury (VILI) was an important factor. 2, low tidal volume mechanical ventilation on lung tissue HMGB1 expression was less affected, no obvious damage to normal lung tissue. 3 injection Xuebijing to a certain extent, can inhibit the lung tissue HMGB1 mRNA and protein expression induced acute lung injury have some preventive effect on mechanical ventilation.
