Abstract


Objective: This issue through the detection of angiotensin II (Ang II)-induced endothelial progenitor cells (EPCs) apoptosis apoptosis and the transcription factor FoxO4 expression changes, to explore the mechanism of the FoxO4 participate in Ang II-induced apoptosis of EPCs. Methods: wife umbilical cord blood collected 2 hours after delivery, extracted from mononuclear cells, vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) induced by 7 days of culture identified as EPCs in the medium. Grouping of samples based on the different intervention methods: (1) Ang Ⅱ group (n = 6), at concentrations 10-6 mol / L of Ang II-induced EPCs apoptosis;, Ang Ⅱ chlorine losartan potassium PD123319 group (n = 6), joined AT receptor antagonism The agent 10-6mol / L losartan potassium and 10-6mol / L PD123319 intervention After 1 hour, added at a concentration of 10-6mol / L Ang II; (3) the establishment of the blank control group (n = 6), the specimen is not any intervention. Results: (1) Ang II group apoptosis rate highest, compared with the control group was significantly higher (P lt; 0.05), followed by the Ang Ⅱ losartan potassium PD123319 rate of apoptosis, apoptosis rate differences compared with the control group was not statistically significant (P gt; 0.05) Ang Ⅱ can induce apoptosis of EPCs, this process of apoptosis can be AT receptor antagonist to reduce EPCs apoptosis. ② immunohistochemistry results showed that Ang Ⅱ group FoxO4 expression of the highest positive rate FoxO4 expression in the control group, the lowest positive rate of Ang II Losartan Potassium the PD123319 group of FoxO4 expression rate is between Ang Ⅱ group the FoxO4 positive rate with the control group significant difference (P lt; 0.05), Ang II chloride losartan potassium PD123319 group FoxO4 positive rate with the blank control group comparison no significant difference (P gt; 0.05) ③ transcription factor FoxO4 positive expression group EPCs cell apoptosis rate changes consistent. Conclusion: ① concentration of 10-6 mol / L Ang Ⅱ induced apoptosis of endothelial progenitor cells, Ang II-induced apoptosis of endothelial progenitor cells can be receptor antagonist AT1 and AT2 receptor antagonist; (2) transcription factor FoxO4 The positive expression rate of apoptosis rate consistent with the change, suggesting that FoxO4 transcription factor involved in high concentrations (10-6 mol / L), Ang II-induced apoptosis of EPCs process.
