Background: At present, China is still the countries most affected by silicosis hazard, exposure to silica dust in the number of new silicosis patients currently suffering from silicosis number of ranking first in the world. Silicosis pathogenesis is still not yet clear, scholars have proposed a variety of hypotheses, such as inflammation theory, immune theory, chemical poisoning theory, mechanical stimulus doctrine, silicate polymerization doctrine, surfactant theory, although each theory has its own reasonable some respects, but are difficult to explain satisfactorily the pathogenic process. In recent years, the cytokine network in the pathogenesis of silicosis become a hot spot. The body, Th1-type cytokines and Th2-type cytokines affect each other and interact to form the balance of cytokine network, maintain the body's normal physiological conditions. Once this cytokine network balance is broken, the body will appear abnormal reactions and pathological states. In this study, silicon dioxide (SiO2) rat rats with experimental silicosis model to monitor Th1-type cytokines interferon-gamma (IFN-gamma) and Th2-type cytokines interleukin prime -4 (IL-4) in the lung tissue Expression changes, and to explore the role of Th1-type cytokines and Th2-type cytokines in the process of the formation of silicosis. Nuclear factor-kappa B (NF-kappaB) and transforming growth factor-beta1 (TGF-beta1) has a variety of biological functions. Many studies have shown that NF-kappa B and TGF-beta1 in the pathogenesis of pulmonary fibrosis play an important role by monitoring both the expression in the lung tissue of rats in the silicosis model group and control group in this experiment, investigate the incidence of silicosis possible role in the mechanism. Objective: To clarify the dynamic changes of cytokines in the lung tissue of rats with experimental silicosis disposable after dust; IFN-gamma and IL-4 expression in the lung tissue of rats with experimental silicosis and significance; explore the NF-kappa B and TGF -beta1 in the lung tissue of rats with experimental silicosis in the expression and functional role. Methods: Rat model of silicosis, disposable Dust on 1,7,14,21,28,35 d (days), respectively, after dust three model group and two control rats were sacrificed and lung tissue Production of tissue sections. Immunohistochemical SABC detected by IFN-gamma and IL-4 expression in 18 cases of rat model of experimental silicosis and 12 cases of normal lung tissue of rats, IFN-gamma and IL-4 expression quantitative image analysis system analysis; immunohistochemical SP method NF-kappa B and TGF-beta1 expression in 18 cases of rat model of experimental silicosis and 12 cases of normal lung tissues of rats, the expression of NF-kappaB and TGF-beta1 quantitative analysis . Results: Compared with the control group, a one-time after dust 1,7,14,21 d, IFN-gamma expression in the lung tissue of rats in the model group significantly increased (P lt; 0.05), IL-4 expression was significantly reduced (P lt; 0.05); 28, 35 days after dust IFN-gamma expression in the lung tissue of rats in the model group weakened, there was no significant difference with the control group (P GT; 0.05); IL-4 expression significantly increased the amount was significantly higher (P lt; 0.05). , 7, 14 d after exposed to silica, silicosis and lung tissues of rats NF-kappaB and TGF-beta1 expression with the control group significantly increased (P lt; 0.05); levels of expression of both 21d decline, 28d both expression level close to the control group compared with the control group, no significant difference (P GT; 0.05) To the 35d, the NF-B expression significantly reduced, compared with the control group, there were significant differences (P lt; 0.05), while no significant difference between the expression of TGF-beta1 with the control group (P gt; 0.05) Conclusion: In early experiments (1-21D), the Th1-type cytokines IFN-gamma expression was significantly increased, indicating that silicosis rats Th1-type cytokine-mediated cellular immune response; experimental late (28-35d) Th2 cytokines IL-4 expression was significantly enhanced IFN-gamma expression weakened, silicosis rats in vivo cellular immune response began to decrease, the humoral immune response mediated by Th2-type cytokines began to increase. Silicosis model early (1-14d), NF-kappaB and TGF-beta1 expression were significantly enhanced, indicating that the two may be involved in the formation of early silicosis fibrosis.
