Abstract

Test meat processing abandoned pig pancreas research material, isopropanol precipitation trypsin method and the the chitosan flocculation trypsin law are two ways to study the extraction of trypsin, for the industrial production of trypsin methods and The process parameters provide a reference. Test on the first preferred activator and protective agent, further use of the single factor and orthogonal experiments, study of the trypsin extract the main factors and process parameters identified two ways to produce trypsin process The optimal process parameters, the last of the trypsin powder biological properties studies by pancreatin vitality, as well as the determination of the physical and chemical indicators, the results are in line with the Chinese Pharmacopoeia standards. The results are as follows: 1. Determine the trypsin method isopropanol precipitation trypsin law and chitosan precipitation process: raw frozen pancreas minced after pancreas, activate, extraction, filtration, sedimentation, defatted and dried under vacuum to give pancreatin. Plus extractant extracted through single factor test, select the first add activator to activate as the best method of activation; CaCl2-trypsin powder is selected for optimal activator combination. The selected starch, maltose as a protective agent. Isopropanol precipitation method: a plurality of sets of single factor test to elect the best factors affecting the extraction of the pancreatin is isopropanol concentration, the pH, the extraction time, the activation time, the activation temperature, precipitated alcohol degrees, and by L 18 (3 7 ) orthogonal test, screening out the optimal parameters to the tryptase activity as indicators: isopropyl alcohol concentration of 15%, pH6.5, extracted 3h activation time 8h, activation temperature 8 ° C to precipitate alcohol of 60%. The resulting trypsin yield 10.79%, of trypsin activity 4791 U / g, pancreatic amylase activity 57806 U / g pancreatic lipase activity 45 061 U / g, 4.61% Loss on drying, the fat content of 18.19mg / g, in line with the Chinese Pharmacopoeia. 4. Chitosan flocculation law extraction trypsin: elect affect trypsin extract the best single factor test for alcohol concentration, extraction time, pH, chitosan dosage, and through L 9 (3 4 ) orthogonal experiment, the trypsin activity indicators, filter out the optimal parameters: 10% ethanol alcohol, extraction time 6h, pH 4, chitosan The dosage is 0.1%. The resulting trypsin yield 9.24% trypsin vitality 3703 U / g, pancreatic amylase activity 54285 U / g pancreatic lipase activity 44 552 U / g, and 4.86% Loss on drying, the fat content of 17.58mg / g, in line with the Chinese Pharmacopoeia. 5 pancreatin biological characteristics: optimum temperature of porcine pancreatin 40 ℃, found by thermal stability studies, the trypsin activity gradually decreased with increasing time, the higher the temperature, the faster deactivation of the trypsin. The pancreatin optimum pH value of 8, pH stability study found that trypsin activity gradually decreased as the pH rises higher, pH, the faster the inactivation of trypsin. By the of Ca 2 and EDTA trypsin activity found, Ca 2 had no effect on the activity of trypsin EDTA on the strong inhibition of protease activity . In summary, isopropanol precipitation the trypsin method chitosan precipitation trypsin law have the process is simple, short production cycle, the system of trypsin high quality, isopropanol precipitation produced trypsin higher yield, three-enzyme activity is higher than the latter, there is a good prospect. Therefore, the use of advanced separation of biological engineering technology for the production of trypsin enzyme yield and vigor, biochemical pharmaceutical industry and the comprehensive utilization of animal by-products has a positive meaning.
