Abstract

The green tide large marine green algae occur frequently at home and abroad in recent decades fulminant growth and floating growth of harmful ecological anomalies, mainly from the original solid growth transformation occurs in the estuary, dense inner bay, lagoon and city the coast, the scale and types of outbreaks vary from place to place. Forming green tide algae the Ulva Branch (Ulvaceae) green algae the seaweed Ulva genera (Ulva). In 2008, the the floating the Enteromorpha Tailv tide outbreak of the Yellow Sea in south-central cause of the concern of the national marine green tides; 2009, the Qingdao coastal areas and several large accumulation of floating green algae Ulva genus of marine ecosystem is broken environment caused by the negative impact. In order to find the green tide occurred, and further to achieve the purpose of prevention, care green tide, you first need to do accurate identification of the cause green tide species. In this study, the work is mainly floating growth of green algae in Qingdao coastal areas, species identification, and to determine the source of the most fundamental basis for subsequent analysis and governance. In this study, a digoxigenin-labeled molecular probe technology, the establishment of the Yellow Sea green tide Enteromorpha (Ulva prolifera) rapid identification techniques, DNA extraction, amplification of specific sequences of the test sample, the dual-probe hybridization ultimately achieve rapid accurately identified the purpose of Enteromorpha entire appraisal completed within 24h-36h. This study through amplification and analysis of 18S-28S ITS sequences (internal transcribed spacer 5S ribosomal RNA tandem repeat (5S rDNA spacer) successfully identified from May 2009 to July Qingdao formation within the Bay floating green algae algal blooms with normal solid with the growth of the long stone pertusa species as long stone pertusa (Ulva linza), and to estimate the biomass. discovered form of floating long stone Brasenia the occurrence of a significant change in the amplification and analysis of rbcL (ribulose-1 5-bisphosphate carboxylase / oxygenase large subunit) sequence, found that since July 2009, Qingdao grown within the Bay floating green algae Ulva pertusa (Ulva pertusa) and hard Ulva (Ulva rigida) majority of Ulva pertusa through on-site investigation at the same time, as well as the analysis of the three green algae morphological characteristics that the three green algae do not have the form of the long-range transportation flowing conditions. So, Qingdao sea area in 2009, a large number of floating growth of long Ulva, Ulva pertusa and hard Ulva species are Qingdao local growth, rather than to drift from the open sea. microstructure observation results show that the growth characteristics of the floating growth of green algae cell wall thickening, speculated that their in order to adapt to the changes in survival potential environmental changes for the present study and the application of more standardized the species classification techniques - DNA barcode technology (DNA barcoding), the study of ITS sequences as Ulva Division algae DNA barcode do analysis found ITS sequences in some species of intraspecific genetic distance greater than species interspecific genetic distance, the ITS sequence as the DNA barcode sequence has not yet been reached, but from established phylogenetic tree can be seen, ITS sequences can a good reference for the the species Ulva Division Classification.
