Abstract
Abiotic stresses such as drought, salinity and low temperature is the most important factor limiting crop yield and quality , to cultivate strong resistance of new varieties of crops have important significance for improving agricultural productivity and conserve resources . Transgenic breeding is an effective way to improve crop resistance . Some transcription factor plays an important role in the regulation of plant stress reaction , due to a transcription factor to regulate the expression of genes related to resilience in the crop resilience breeding through genetic transformation of a transcription factor gene can be integrated to improve crop resistance AP2/EREBP of a class of transcription factors unique to plants , and many of the family members play an important role in plant defense reaction . HRD transcription factor is a newly discovered plants in response to drought stress regulatory protein , is a transcription factor AP2/ERF-like subfamily of plant water use efficiency under drought conditions . Due to the HRD gene intron , this study directly cloned from Arabidopsis genome by PCR method HRD gene sequencing correct . To strong expression vector pBIN438 as the skeleton by restriction enzyme digestion , the connection was carrying HRD gene plant expression vector pBIN- HRD , transformed by Agrobacterium . Regeneration of genetically HRD gene containing the plant expression vector pBIN-HRD tumefaciens GV3101 strains of the tomato infection , after co-culture , the induction of adventitious buds , rooting transplanting seedlings . Regenerated plants for the purpose of gene PCR and RT-PCR analysis , the results suggest that the Arabidopsis thaliana the HRD gene has been integrated into the tomato genome and expressed in some plants . Analysis of the positive transgenic tomato chlorophyll content , leaf water retention test and PEG - induced drought analysis , the results showed that the HRD gene in a certain extent, can improve drought resistance of tomato . This experiment also transformed tobacco regeneration of transgenic tobacco plants . HRD gene cloned from Arabidopsis using Agrobacterium-mediated transformation HRD gene into the tomato and stress resistance of transgenic tomato plants , which have greater resilience for crop breeding research .
