Plants under abiotic stresses will produce a large number of toxic effects of reactive oxygen species, reactive oxygen scavenging system is of great significance for the plant to maintain its normal physiological function. Melon and apples are a favorite fruit, the fruit is sweet and delicious flavor. The new varieties using biotechnology to cultivate high-nutrition, stress tolerance is one of the main objectives of the current breeding. Active oxygen scavenging enzyme activity, thereby enhancing the ability of the plant's resistance to oxidative stress, high stress resistance of new varieties to lay the foundation for breeding; and genetic engineering methods to improve study the metabolic pathways and ascorbic acid - glutathione Gan peptide cycle the foundation. This article main findings are as follows: 1 to Muskmelon varieties 'the West Sweet One' cotyledons and leaves as explants through organogenesis pathway to form adventitious buds, to explore different hormone combinations, leaf age, explants placed , with or without dark culture conditions for its regeneration. Results show that ,5-7 d-old cotyledons block MS 6-BA0.8 mg / L of the induction medium, growth blade around two weeks after subculture in MS 6-BA1.0 mg / L induction medium contact with the medium, on the abaxial face down directly into the normal light cycle culture can be 100% recycled. MS 6-BA0.05 mg / L adventitious shoot elongation medium, the differentiation of adventitious buds (Cong) elongation grew up. 1/2MS IAA0.2 mg / L rootless seedlings rooting medium hair root, rooting rate of up to 100%. Constructed dehydroascorbate reductase gene (DHAR, serial numbers DQ322706, cloned from 'Gala' apple leaves) plant expression binary vector and introduced into Agrobacterium. 3 In the to establish the cotyledons regeneration system and construction of expression vectors based on 'the West Sweet One' melon cotyledons as explants, Agrobacterium mediated method DHAR gene transferred to the melon. Hygromycin resistant plants and PCR-positive plants as a preliminary determination of the transgenic plants to study the impact of conversion factors. Cotyledons after 2 d pre-culture, OD600 value of 0.6 of the bacterium infection 8 min 3 d of culture, the the hygromycin pressure of 4 mg / L the the antibacterial prime cefotaxime concentration of 500 mg / L conditions, the conversion rate is the highest positive transformed plants initially identified 17 resistant plants after PCR assay. 4 of melon positive transformed plants in six lines ascorbic acid levels were measured, the total AsA content increased significantly, the highest increase in more than 2 times, but most of the strains redox ability did not significantly change. These preliminary findings indicate that Apple DHAR gene was successfully transferred to the melon and DHAR overexpression can improve AsA content. 5 of the previous studies of this laboratory 'Changfu 2' Apple regeneration system and genetic transformation system, DHAR gene transformation 'positive transformed plants Changfu 2' Apple, after PCR assay initially identified a resistant plants .
