Abstract


The iron Dendrobium (Dendrobium candidum Wall.ex Lindl.) The protocorm Iduced a callus embryogenic callus dark subcultured 2 times as experimental materials to study a variety of sugar of the more injured tissue growth and biochemical characteristics. The main results are as follows: the embryogenic callus subculture were used to study the effects of sucrose, fructose, trehalose, maltose and lactose callus growth. The results showed that: sucrose, fructose, trehalose, is conducive to the growth of the callus, the optimum concentration thereof is 3%, respectively, 2%, 3%; maltose second, the optimum concentration of 2%; lactose is most detrimental to the more injured tissue, the optimum concentration of 0.5%; fructose and sucrose on callus growth start time earlier, while trehalose is late, longer duration of trehalose and fructose callus growth, lactose, sucrose, and maltose shorter duration of action; various sugar callus proliferation rate almost all first rose to a peak and then decline, callus slow growth at lower concentrations, whereas at higher concentrations, but also inhibited the growth of the callus. 2 in the above basis, trehalose, sucrose, and lactose soluble sugar content in callus. The results show that: the soluble sugar content within the calli with the three kinds of sugar increases with increasing glucose concentration of the medium, the calli cultured in the medium of the three kinds of sugar has an in vivo, in vitro sugar chemotactic balanced concentration, respectively 6%, 3%, 0.5%, the growth of callus in the sucrose and lactose medium vivo sugar tends to balance concentration callus growth optimum concentration exactly consistent, while in trehalose medium grown on both concentrations of the callus not consistent. 3 when trehalose, sucrose, lactose, in a callus growth rate reached a maximum, the callus three isozymes EST, SOD, AMY influence. The results are as follows: in addition to lactose medium callus EST isozymes no significant difference in the sucrose and trehalose medium has a larger change in the emergence of new enzymes with higher sugar concentration , indicating that there may be a new gene expression, and at a concentration of 5% and 9%, respectively enzyme band deeper enzyme bands are shallower, but at a concentration of 3%, with the front of the callus proliferation optimum balance the concentration of the concentration and in vivo, the outer sugar reached consistent; the SOD enzymes spectra of the callus on three kinds of sugar medium, at high concentrations, there will be the emergence of new enzymes with the three control with increasing time, SOD enzyme with gradually reduced; three kinds of high sugar concentration, amylase enzyme with deep and high soluble sugar content; 3 contrast amylase enzyme with deeper and increase over time, amylase enzyme band also will be deepened, and soluble sugar content is less and less. Obtained sugars conducive to of Dendrobium candidum callus growth and its optimal concentration of soluble sugar content in callus of three kinds of sugar, and by the cultured callus EST on three kinds of sugar, SOD AMY isoenzyme analysis shows that the medium sugar can regulate gene expression in embryogenic callus development process.
