Abstract


One of the most important environmental factor alone plant growth and development , is now widely used in the form of plant tissue culture process , growth control photomorphogenesis its reaction mechanism , a large number of research results show that a variety of light quality on the form of plant somatic embryogenesis , organ differentiation , biochemical indicators have to a certain extent . In this experiment, Dendrobium officinale induced embryogenic callus raw materials , red NO.1 red NO.2 , green, blue , white , dark six kinds of light quality processing , morphological level change observed on some of the morphological characteristics of the process of somatic embryo development , and the peroxidase enzyme ( POD ) , esterase ( EST) isozymes of cytochrome oxidase (COD) spectrum analysis . Morphological study of of Dendrobium candidum body in a variety of light quality embryo , in the 42d of the culture period , light quality on somatic embryogenesis of Dendrobium candidum growth of varying degrees of role : Blu-ray > white > red NO.1> red NO.2> green > darkness ; each growth period under different light quality per gram fresh weight growth rate of the order : white > Blu-ray > red NO.1 > green > red NO.2> dark . Of of Dendrobium candidum somatic embryos color change observed , green and white light enables organizations to block quickly turns green , followed by blue light . The experimental conditions of light quality POD EST COD isozyme spectrum of the enzyme with the number and intensity of the enzyme with the enzyme with Rf value analysis , and found a common enzyme with each light quality : POD isozymes with Rf P16 , Rf , P20 < / sub > ; EST isoenzymes with Rf E7 the Rf E11 ; COD isozymes with Rf C13 , Rf C15 . Also found in certain light quality unique enzyme with 14 : NO.1 the next red light three ; 1 each green and blue : three white light ; dark 6 . The combination of the enzyme with the characteristics and morphological characteristics , we can isozymes indicate a preliminary understanding of Dendrobium candidum somatic embryos under different light quality during development .
