Abstract


Background is widely neuropathic pain and inflammatory pain on brain-derived neurotrophic factor (BDNF) has been preliminarily confirmed that BDNF is extremely important neurotransmitters in the generation and maintenance of neuropathic pain and inflammatory pain . Intervention for spinal cord BDNF levels can play mitigate the effect hyperalgesia . Produce BDNF in cutting pain to maintain as well as cutting after the hyperalgesia process in the role of , however , is still poorly understood. Cutting widespread pain in life, human suffering , and solve the problem of pain after surgery is still not satisfied . These have prompted us to do a more in-depth study of the mechanism of cutting pain . This experiment is trying to explore the role of the dorsal horn of the spinal cord and dorsal root ganglia (DRG) in BDNF cutting pain . Under isoflurane anesthesia longitudinal cutting the hind foot , taken in different periods after surgery of the spinal cord and DRG observed changes in the spinal cord dorsal horn and DRG level and expression of positioning . Immunofluorescence double labeling method and immunohistochemistry chemical methods were applied to detect BDNF levels . Observe the sciatic nerve block after lidocaine spinal dorsal horn and BDNF levels within DRG . One hour before cutting intrathecal and intraperitoneal injection of anti- the BDNF serum , to observe the behavior of the rats hind cutting histological changes . Results after rat foot cutting led to the ipsilateral DRG and lumbar spinal cord BDNF levels increase , thoracic did not change significantly . Spinal cord BDNF levels raised mainly located in neurons and not astrocytes or microglia cells . Before cutting applications Bureau anesthetic lidocaine sciatic nerve block ipsilateral DRG and lumbar spinal cord BDNF levels increase . Intrathecal injection of anti- the BDNF serum can significantly inhibit the cutting - induced hyperalgesia . Intraperitoneal injection of anti-BDNF serum no such effect . Conclusion The study showed that cutting led to the ipsilateral DRG and lumbar spinal cord dorsal horn BDNF levels increase afferent somatosensory closely related . Increased BDNF actively involved in cutting -induced hyperalgesia .
