Objective : Understanding of arsenic trioxide on lung adenocarcinoma cell line A549 cell proliferation, cell cycle , apoptosis, and cell survivin, cylin D1 gene expression . Method : different concentrations of arsenic trioxide in A549 cells after different time of cell proliferation activity , cell cycle , apoptosis and cell of survivin , cyclin D1 gene expression changes detected . Results : slightly higher concentrations (> 2μmol / L) Arsenic trioxide can inhibit the proliferation of A549 cells in a dose- and time-dependent manner . Reverse transcriptase polymerase chain reaction (RT-PCR) detection of survivin of cyclin D1 gene transcription level and cell climbing film Immunohistochemical detection of survivin , the relative intensity of the protein expression of cyclin D1 gene , discovered the the arsenic trioxide role of survivin and cyclin D1 gene expression compared with the control group was significantly lower (P <0.05). Arsenic trioxide inhibition of A549 cells from G1 to S phase concluded : Arsenic trioxide can effectively inhibit proliferation of A549 , A549 cells induced apoptosis , inhibition of cell G1 into the S phase , and a positive correlation with arsenic trioxide dose and duration of action , this process may be suppressed and arsenic trioxide survivin, cyclin D1 expression .
