Background: In recent years, the worldwide cancer incidence, mortality rates have continued to rise. One of the main methods of cancer treatment with chemotherapy, but prone to drug resistance. Thus targeted therapy become hot research today, these ideal endostatin. Was first proposed in 1971 Follunan the tumor dependent angiogenesis doctrine, that the majority of tumor growth need of new blood vessels to supply oxygen and nutrients, and clear metabolites. The most solid tumors in the start-up phase can grow to about 2 mm, and no new blood vessel formation and non-proliferation state for years, and the possibility of deterioration in the period. In subsequent tumor metastasis study also found that, when the tumor within the small diameter of metastases in vascular angiogenesis pre-metastatic foci within the tumor cell proliferation and apoptosis rate in equilibrium, micrometastases in the long-term period of dormancy, and once the tumor vascular system in the formation of metastases, tumor cell proliferation and apoptosis balance broken, and the rapid increase of the tumor in the short term. Therefore, inhibition of angiogenesis far-reaching significance and broad prospects for the treatment of tumors. Endostatin (Endostatin) Harvard University in 1997, O'Reilly found a potent angiogenesis inhibitor. Can inhibit vascular endothelial cell proliferation, induce apoptosis and inhibit tumor formation of new blood vessels. Since the target cells is the the genotype relative stability of vascular endothelial cells, rarely mutate, easy to induce resistance. Objective: To observe the growth of lewis lung cancer cell lines after different doses of endostatin intervention, apoptosis resistance protein P170 expression. Endostatin tumor cells. Methods: Using a different dose groups of endostatin lewis lung cancer cell lines (LLC) to intervene and: lewis lung cancer cell growth inhibition assay. Application of flow cytometry to detect apoptosis. Resistance proteins P170 expression by flow cytometry. Results: Endostatin can inhibit the growth of lewis lung cancer cells, its effects and its role is proportional to the time; endostatin lewis lung cancer cells can induce apoptosis, the apoptotic rate was positively correlated with the dose and duration of action to a certain extent; application different concentrations of endostatin treatment LLC were not reflected in its expression of the resistance protein P170. Conclusion: Endostatin can induce apoptosis in tumor cells, its dose proportional to their degree within a certain range. Endostatin can inhibit the growth of tumor cells, a significant time-dependent effect. Did not find the endostatin to affect resistance protein P170 expression.
