Objective To observe the basic fibroblast growth factor (bFGF) on cultured rat intervertebral disc cells and matrix metabolism. Methods Rat intervertebral disc cell culture system in vitro rat intervertebral disc cells in monolayer culture , take the 2nd generation grew well cell experiments , the use of anti-Fas antibody-induced apoptosis in different concentrations (10,100,1000 μg / L) alkaline fibroblast growth Factor on intervertebral disc cell apoptosis and metabolic processes , the use of flow cytometry -PI assay disc labeled apoptosis ; method using the chloramine -T and DMB colorimetric cell culture medium were detected hydroxyl prolyl acid and proteoglycan content . Results ① different concentrations of basic fibroblast growth factor (10,100,1000 μg / L) disc apoptosis rate (87.62 ± 11.06), (53.30 ± 9.23), (16.75 ± 4.21)% compared with the control group, there were significant differences significance (P <0.01). ② With the basic fibroblast growth factor concentration increased (10,100,1000 μg / L), hydroxyproline content , proteoglycan content increased significantly , [ hydroxyproline content (6.71 ± 0.33), (9.12 ± 0.41) , (11.58 ± 0.12) μg / L, P <0.01; proteoglycan content (23.21 ± 2.87), (32.45 ± 5.23), (37.18 ± 3.22) μg / L, P <0.01]. Basic fibroblast growth factor positive correlation between the concentration (ra = 0.972, P <0.01: rb = 0.907P <0.01). Conclusion of basic fibroblast growth factor can inhibit apoptosis in vitro intervertebral disc , intervertebral disc cells promote collagen and proteoglycan matrix synthesis.
