Abstract


Objective: through different doses of recombinant human insulin-like growth factor - Ⅰ (rh IGF-I) to stimulate human osteoblast-like cells MG-63, to explore the insulin-like growth factor - I on osteoblasts vascular endothelial growth factor (VEGF) expression and regularity . Methods: The human osteoblast-like cell line in vitro cell culture , using reverse transcription polymerase chain reaction (RT-PCR) assay different concentrations of rh IGF -I , and the different duration of action of human osteoblast-like cells VEGF mRNA expression impact and the law . The results : (1) RT-PCR detected mRNA expression of varying degrees of MG-63 cell VEGF and internal reference . ( 2) Semi-quantitative analysis showed that different concentrations of IGF-I (10,20,50,100 ng / mL) in 24 hours when intervention MG-63 cells , VEGF mRNA expression levels compared 0ng/mL significantly enhanced by the internal reference correction were approximately 0ng/mL 2.2 times , 3.26 times and 4.37 times , 4.56 times , were statistically significant (P <0.05). ( 3 ) semi-quantitative analysis showed that , when rh IGF-I concentrations between 0 ~ 50ng/ml human osteoblast-like cells VEGF mRNA expression levels significantly increasing relationship and pairwise compared were statistically significant (P <0.05). (4 ) Semi-quantitative analysis showed that when rh IGF-I concentration of 50ng/ml so , compared with 0ng/ml , the highest expression levels of VEGF mRNA was statistically significant ( P <0.05 ) ; over this concentration (50ng / ml), compared with 50ng/ml increased expression of VEGF obvious trend was not statistically significant ( P > 0.05 ) . ( 5 ) semi- quantitative analysis , the application of a final concentration of rh IGF-I (50ng/ml) stimulation of human osteoblasts , the different duration of action of VEGF mRNA expression was significantly enhanced in the peak time 24h , was statistically significant ( P <0.05). Conclusion: VEGF expression in MG-63 cells ; rh IGF -I in vitro stimulation of human osteoblast-like cells MG-63 VEGF expression was a certain amount of time and concentration-dependent relationship .
