Abstract


Objective: To examine the histological change of different layers and stenosis of reference vessels after balloon injury in rabbits. Compare the reference and injury vessels after balloon injury to infer possible mechanism of reconstruction of reference vessels.Methods: Eighteen New Zealand rabbits were given high fat diet for 1 month and randomized to 2 groups, the control group were given high fat diet+ sham operation, operation group: given high fat diet and underwent endothelium injury. The operation group was used to establish balloon injury models in rabbits. Killed all rabbits and got the reference and injured vessels, two and four weeks after operation. The artery samples were fixed with 10% formalin, embedded in paraffin. They were separated and fixed for pathological study. The artery segment was cross-sectioned at 4μm intervals, stained with hematoxylin and eosin (HE) and the other immunohistachemical method. Observe the change of different layers of vessel wall, especially the change of PCNA. Observe the reconstitution of reference and target vessels by histology morphometric analysis.Results: Balloon injury models were established successfully. After procedure, the reference vessels can also observe hyperplasia The tunica intima was thickened, proliferation of VSMCs and fibrous tissue were obvious. The hyperplasia of reference vessels were significantly different to the control group, and there was a significant linear correlation between the reference and target vessels.Conclusion: Through the experiment, we can conclude that the reference vessels also confront hyperplasia after the balloon injury of target vessels, and have a direct correlation to each. So when valuing the reconstruction of target vessels, we must consider that the reference vessels may also have hyperplasia, the evaluation criterion may be adjusted, which needs advanced researches.
