Abstract


Objective: phage amplified biologically the law (PhaB method) direct detection of Mycobacterium tuberculosis in sputum specimens (MTB resistance to isoniazid and rifampicin, in order to establish a high specificity and sensitivity of the MTB drugs sensitive test method: Select the initial clinical diagnosis of drug-resistant tuberculosis 40 in sputum of patients use PhaB law direct detection of MTB in sputum samples of isoniazid and rifampicin resistance, while using the absolute concentration method detection isoniazid and rifampicin resistance, to compare the two results. Results: 40 patients the sputum samples PhaB law and absolute concentration method isoniazid resistance test results were positive in 22 cases, 11 cases were negative, the two laws do not match the seven cases. criteria for judging the absolute concentration method test results, isoniazid resistance the PhaB method detection sensitivity, specificity, positive predictive value, negative predictive value and accuracy were 78.57% (22 / 28), 91.67% (11/12), 95.65% (22/23), 64.71% (11/17), 82.50% (33/40); Kappa test, k = 0.63 P = 0.00 lt; 0.05 Description the PhaB and absolute concentration method test results are in good agreement. the 40 patients the sputum samples PhaB law and absolute concentration method rifampin resistance test results were positive in 25 cases, seven cases were negative, the two laws do not match eight cases . absolute concentration method test results as criteria for judging, PhaB law rifampin resistance detection sensitivity, specificity, positive predictive value, negative predictive value and accuracy were 78.13% (25/32), 87.50% (7/8), 96.15% (25/26), 50.00% (7/14), 80.00% (32/40); Kappa test, k = 0.51, P = 0.00 lt; 0.05 Description PhaB law with absolute concentration method The test results are in good agreement. 40 cases patient sputum samples PhaB law isoniazid and rifampin resistance detection sensitivity, specificity, positive predictive value, negative predictive value, and accuracy of the comparison, P are gt; 0.05, the difference was not statistically significant. Conclusions: PhaB assay method to directly detect sputum samples MTB has a high sensitivity, specificity and accuracy of isoniazid and rifampicin resistance, and absolute concentration method good consistency. PhaB assay method for different drug resistance testing sensitivity, specificity, positive predictive value, negative predictive value, no difference in accuracy. PhaB assay method only takes 3 days, simple and fast, does not require special equipment , low-cost, rapid screening method can be used as MTB resistance testing.
