Abstract


Major histocompatibility complex (major histocompatibility complex, MHC) Ⅱ molecules involved in antigen presentation is the main material, mainly expressed in the immune presenting cells (APC), in inducing the immune response plays an important role. Studies have shown, MHC Ⅱ antigen in the intestinal villus epithelial cells, airway epithelial cells were MHC Ⅱ antigen expression, suggesting that these cells through the process of MHC Ⅱ antigen presentation pathway. Currently on the endometrium is also has a non-professional antigen-presenting cells little studied. In order to investigate the MHC Ⅱ expression in mouse endometrium and variation, testing immunohistochemical staining to detect endometrial tissue in mice of different ages and in vitro cultured mouse endometrial cells is correlated with changes Meanwhile, the determination of the extracellular matrix (extracellular matrix, ECM) cultured on plastic, estradiol and progesterone hormone stimulation, the virus stimulated lymphocyte conditioned medium and other factors on cultured mouse endometrial cells expression of MHC Ⅱ. Obtained the following results: 1. Tested 10 different days (born 5 d, 15 d, 30 d, 60 d, pregnancy, 1 d, pregnancy, 4 d, pregnancy 6 d, pregnancy 9 d, pregnancy 15 d, pregnancy 18 d) mouse uterus MHC Ⅱ expression levels. The results show that the detected age endometrial stromal and epithelial cells express MHC Ⅱ were born after 5 d, 15 d, 30 d, the first 60 days, as age increases, followed by enhanced expression of the difference was significant (P lt; 0.05). The first 60 days, MHC Ⅱ expression of the strongest, but the expression of MHC Ⅱ weakened into pregnancy. During pregnancy, pregnancy 1 d, pregnancy, 4 d, pregnancy 6 d, 9 d of pregnancy among these groups of expression of MHC Ⅱ no significant difference. The pregnancy 18 d and other groups were significantly different (P lt; 0.05), MHC Ⅱ expression of the weakest. 2 cultured primary mouse endometrial stromal cells and epithelial cells in culture 2-4 days MHC Ⅱ expression was positive, with the incubation time, MHC Ⅱ expression gradually weakened, to the first five days, MHC Ⅱ tested negative . 3 cultured on ECM gel covered with mouse endometrial stromal cells and epithelial cells in culture in the first five days could be detected in the expression of MHC Ⅱ. Add estradiol and progesterone on the expression of MHC Ⅱ cells had no significant effect. 4 of 5 days in vitro mouse endometrial stromal cells and epithelial cells culture medium stimulated lymphocyte virus conditioned medium can induce the expression of MHC Ⅱ. To illustrate these results in mouse endometrium local immune cells - the role of endocrine regulation provides experimental evidence.
