Mandibular defects caused by trauma, infection, tumors, and congenital diseases are very common, major repair method for mandibular defects with autogenous bone graft, which is most common as in the iliac bone donor. Embryological studies have shown that most craniofacial bone occurs in the cranial neural crest source of ectodermal mesenchymal trunk limbs from the mesoderm; intramembranous osteogenesis maxillary bone, iliac, compared with cartilage within osteoblasts. A large number of clinical and animal experiments show that the truncated nature mandibular defects with mandibular donor for the body of a higher success rate than other parts of the bone, mainly as transplant donor survival rate is higher, lower absorption rate forward. These existing clinical, laboratory, biological studies suggest that bone progenitor cells in different parts of the bone marrow may have their own biological characteristics; cytology model on the repair of bone tissue from the long bone of the jaw source of bone marrow cells The little targeted research. To this end, we have adopted the rat mandible with iliac bone marrow stromal stem cells (Bone Marrow StromalCells, BMSCs) were cultured in vitro, by inducing conditions, observed two different sources of BMSCs in cell proliferation and colony formation mineralization formation and differentiation of adipogenic differentiation into bone, whether there are differences, the hope is to further explore the mandible in the event of repair whether its unique biological mechanisms provide the basis and reference. The first part of the rat mandible and iliac source of bone marrow stromal cells cultured in vitro Objective: To explore the use of whole bone marrow cells adherent culture two points of BMSCs were cultured from rat mandible with iliac bone marrow jawbone and The biological characteristics of the ilium sources BMSCs sites difference whether cell model. Methods: 150 g ± 10 g of 4 to 6-week-old male SD rats were first dissecting the mandible and ilium, under sterile conditions, the medium out of the bone marrow cells into cell suspension by whole bone marrow adherence method cultured cells, phase contrast microscope to observe the situation and characteristics of cell growth. Results: The primary cells 24h after inoculation microscope the bottom adherent cells, medium was changed for the first time removal of most of the peripheral blood cells were seen scattered in the spindle cell aggregation; With the medium was changed repeatedly, bottle nonadherent cells gradually being Clear cell growth and the formation of a typical fibroblast-like - colony-forming units (colony-forming unit-fibroblast, CFU-F). Cells were first passaged takes about two weeks or so, the passage cells before 6th generation (ilium) - 9th generation (mandible) relatively strong cell proliferation, cell viability after waning different differentiation tendencies, some cells intracellular vacuoles and granular material in the pulp, and ultimately death. Conclusion: The whole bone marrow cells adherent culture to cultivate meet the requirements of the experimental BMSCs. A Comparative Study of the second part of the source of rat mandible and iliac bone marrow stromal stem cells in vitro biological characteristics Objective: To investigate whether there are differences rat mandible with iliac sources BMSCs in vitro biological characteristics in vitro environment, for further in vivo studies provide basis. Methods: The second generation of the normal growth of the mandible and iliac BMSCs, respectively, from the activity of cell proliferation, induction into the differences in bone mineralization and adipogenic ability compare both in vitro biological characteristics. Results: the mandible sources BMSCs compared with the ilium, in low concentration vaccination conditions under stronger proliferation, induction into the bone higher alkaline phosphatase positive performance mineralization induced formation of calcium nodules ; iliac sources BMSCs differentiate into adipogenic conditions more lipid. Conclusion: the mandible, and iliac source BMSCs in vitro with different biological characteristics, prompt the mandible sources BMSCs may have its unique biological characteristics, provided the experimental basis and reference for further study of the jaw bone lesion regeneration and repair mechanisms.
