Abstract


Object:Reseach the extraction, the qualitative and qualitative analysis of the flavonoids from the seeds of the plant(Garduus crispus L.).Research the protection for mice liver injury by three models and discusses its enzyme, protecting liver, and adjust the immune function, and provides a new pharmacology for further research.Methods:The experimental research method using ethanol extract, ethyl acetate extraction and petroleum ether from the plant and purification methods from seed of the plants and useing uv color, ammonia, Alcl3 reaction, and paper chromatography method appraisal the nature of flavonoid compounds, and by uv spectrophotometry, the flavonoids content is determined.This experiment use mice as experimental object and copy three different animal models information of liver damage through gastricing lavage. Using carbon tetrachloride duplicate the mouse liver injury and using chemical BCG and LPS fatliquoring polysaccharide lipopolysaccharide of mice induced liver damage, and animal model (SRBC) inducing sheep red late to liver damage abnormal reaction mechanism of animal models. Through the mice viscera index, biochemical, ALT levels as well as the AST mouse liver super oxide dismutase SOD activity organization and malondialdehyde MDA content determination, to observe the influence of liver damage with the flavonoids in mice.Results:1. Using the method of ethanol, ethyl acetate extract extraction and purification methods of petroleum ether extract flavonoid and measuring the content of flavonoids compounds,36.6%.Comparied with rutin, use ultraviolet spectrophotometry Garduus crispus L in plant seeds .This method is feasible, simple operation, is a better way of extracting flavonoids substances. The extraction rate of flavonoids is 1.3%, the content of flavonoids is 36.6%,the totle extraction rate of flavonoids from the seeds of the plant is 0.49%.2. Chemical liver injury in comparison with the control model group, the model group was obviously higher than placebo visceral index and ALT and AST were significantly higher, show the carbon tetrachloride (CCL4) copy mice chemical liver damage test model success, Each medicine group of visceral index significantly better model group (p < 0.05), the treatment group than in the model group, the AST ALT decreased significantly (p < 0.05), liver SOD in model group, MDA content increased significantly in model group significantly reduced. Flavonoids from the plant can effectively resist the mice liver damage caused by CCL4.3. The mice liver injury caused by immune cells by injecting BCG + LPS.Dates induced in model group visceral index significantly higher blank group, and ALT and AST were significantly increased, show the autoimmune liver due to LPS and BCG damage caused by the immune cells test model success, All the viscera index is obviously superior model group (p < 0.05), the treatment group than ALT, AST model group decreased significantly (p < 0.05), liver SOD in model group, MDA content increased significantly in model group significantly reduced. Flavonoids can effectively resist the liver damage by mice immunity caused by BCG + LPS.4. The cell autoimmune liver damage caused by SRBC in model group visceral index significantly higher blank group, and ALT and AST were significantly increased, show autoimmune liver damage model due to SRBC to build a successful.All the viscera index is obviously superior model group (P < 0.05), the treatment group than ALT, AST model group decreased significantly (P < 0.05), liver SOD in model group, MDA content increased significantly in model group significantly reduced. Proof of Garduus crispus L,Flavonoids can effectively resist the liver damage by mice immunity caused by SRBC.Conclusion:1. Flavonoids from seeds of the plant were qualitative identified.The extraction rate of flavonoids is 1.3%, the content of flavonoids is 36.6%,the totle extraction rate of flavonoids from the seeds of the plant is 0.49%.2. Flavonoids from seeds of plant Garduus crispus L can effectively resist the mice liver damage caused by CCL4.3. Flavonoids from seeds of plant Garduus crispus L can effectively resist the liver damage by mice immunity caused by BCG + LPS.4. Flavonoids from seeds of plant Garduus crispus L can effectively resist the liver damage by mice immunity caused by SRBC.
