Study of Oridonin on SGC-7901 Cell Proliferation by Inhibitting Cell Cycle Proteins

Abstract


There are many reports on anti-tumor effect of oridonin in our country, suggesting broad-spectrum anti-tumor effect of oridonin. At the same time, It was found that oridonin can inhibit tumor proliferation through the cycle and apoptosis mechanism, but the mechanism of SGC-7901 cell cycle arrest has not been well studied action, so the main subject of this study is to investigate the anti-tumor mechanism of oridonin to SGC-7901 cells in vitro.The growth inhibition of Oridonin on SGC-7901,HepG2,Lovo cells were detected by MTT assay; the effect of oridonin on SGC-7901 cell cycle and a-tubulin were measured through Flow cytometry (FCM) and CLSM respectively. The expression of Cdkl and CyclinBl were analyzed by Western Blotting.The results showed that oridonin had powerful inhibitory action on human gastric cancer SGC-7901, human hepatocellular carcinoma HepG2, human colorectal cancer Lovo cells which cultured in vitro, IC50 values were 15.64,25.50,18.79μM; flow cytometry results showed that the proportion of G0/G1 phase of GC-7901 cells was significantly decreased, whlie the proportion of S phase and G2/M phase was significantly increased by administrating with oridonin for 24 h; the proportion of G0/G1 phase was degraded, but that of G2/M phase was significantly higher by administrating with oridonin for 48 h; laser confocal results showed that, a-tubulin synthesis were blocked, microtubules disappeared, the cell mitotic blocked, after SGC-7901 cells were treated with oridonin;Westem-Blot results showed that the protein expression of Cdkl and CyclinBl significantly reduced with the increasing concentration of oridonin.In summary, oridonin can reduce cycle protein expression of Cdkl,and CyclinBl, inhibit the synthesis of a tubulin, Ihe SGC-7901 cells were blocked in G2/M phase, Finally SGC-7901 cells growth were inhibited.
