Abstract


Objective: To investigate Oridonin (oridonin) preconditioning on rat liver resection autologous transplantation ischemia-reperfusion injury in rats and Nrf2/Keap1-ARE signaling pathways . Methods: Rat liver resection combined with autologous liver transplantation model . A total of 54 male SD rats , according to the preoperative pharmacological preconditioning of different models were randomly divided into sham operation (sham operation group, SO group ) , saline control group (control group, CT group ) and oridonin pretreatment group (oridonin pretreatment group, OP group ) , n = 18 . Were isolated rat liver resection liver autotransplantation after reperfusion or sham surgery group after 2h, 6h, 24h each time point the rats were sacrificed , serum aspartate aminotransferase (AST) level, alanine aminotransferase (ALT); HE staining of liver biopsy for pathological histology ; determination of liver tissue myeloperoxidase (MPO) activity , superoxide dismutase (SOD), glutathione (GSH) activity of glutathione glutathione peroxidase (GSH-Px) activity , Western blot detection of Nrf2, HO-1 protein expression . Results : 1. Compared with SO group , CT group at 2h, 6h and 24h time points : (l) shows a clear liver tissue edema, exudation and neutrophil infiltration ; ( 2 ) liver tissue MPO activity increased (P lt; 0.01), SOD activity decreased (P lt; 0.01), GSH and GSH-Px activity was significantly decreased (P lt; 0.01, P lt; 0.01); (3) serum AST, ALT levels were significantly higher ( P lt; 0.01, P lt; 0.01); (4) liver tissue Nrf2, HO-1 protein expression increased (P lt; 0.01, P lt; 0.01). (2) Compared with the CT group , OP group at 2h, 6h and 24h time points : (l) the degree of liver tissue injury and pathology significantly reduced ; ( 2 ) liver tissue MPO activity was reduced (P lt; 0.05), SOD activity increased (P lt; 0.01), GSH and GSH-Px activity was significantly increased (P lt; 0.01, P lt; 0.01),; (3) serum AST, ALT levels were significantly lower (P lt; 0.01, P lt; 0.01 ) ; ( 4 ) liver tissue Nrf2, HO-l protein were enhanced (P lt; 0.05, P lt; 0.01). Conclusions : 1. Isolated rat liver resection after liver transplantation of autologous often severe hepatic ischemia-reperfusion injury, increased oxygen free radicals are involved in the pathogenesis of one of its principal . 2.oridonin pretreatment reduces rat hepatic resection after liver transplantation of autologous hepatic ischemia-reperfusion injury, which may be related activation Keapl-Nrf2/ARE signaling pathway , increasing phase II enzymes and antioxidant enzyme levels.
