Abstract

In recent years, laser-induced fluorescence spectrum with its real-time, non-toxic side effects and high sensitivity, has been widely used in the field of cancer diagnosis. But academia is not yet cancerous characteristics which fluorescent material identifies organizations form a consistent standard. In addition, the application of this method for tumor diagnosis is mainly concentrated in the examination of the isolated tissue,, Spectral detection based on the organization of the endoscope in the body, these two methods are not provided for surgical procedures peritoneal nodules real-time information, hinder the laser-induced autofluorescence spectroscopy (Laser-induced Autofluorescence LIAF) technology to further promote. Firstly, to build a 405nm laser as the excitation source LIAF system, collected by different biological immune cell therapy, there are different degrees of necrosis nude mouse model of human gastric cancer in vitro tissue LIAF spectrum, monitoring endogenous protoporphyrin IX (protoporphyrin IX, Pp of IX) content of the change, away from the body tissue in the protoporphyrin IX fluorescence peak intensity reduces with the increase in the area of ??tumor necrosis, and nicotinamide adenine dinucleotide (Nicotinamide of Adenine dinucleotide, NADH) 's no obvious change in the law, and the Pp IX of gastric tissue growth state more sensitive identification. Select different human gastric cancer nude mouse model of tumor cell growth time as the research object, the application to build the system, the process of simulated surgical organization of spectrum detection, peritoneum and abdominal tissue in the body spectrum detection. Peritoneal tissue from the group average spectrum form for comparison and found that with the increase of time of the growth of tumor cells, NADH fluorescence peak intensity is gradually reduced, protoporphyrin IX first fluorescent peak intensity increased gradually, indicating that the process of tumor growth, the tissue in the NADH content. gradually reduce gradually increasing the content of porphyrins; hierarchical clustering methods, these organizations are divided into three categories, discriminant analysis method to calculate the discriminant function, to get the overall misclassification rate of 9.6%, which on a normal peritoneal the spectrum misclassification rate of 33.4%, the rate of 6% in the misclassification of the Oncology Group peritoneal spectrum classification diagnosis research on organizational spectrum provides a reference for the application of discriminant analysis. Celiac spectrum collected using principal component analysis method to extract the liver, the spectral characteristics of the three positions in the upper abdomen and lower abdomen and classification. The result found that: the average spectrum of the three types of organizations in the nude mice intraperitoneally forms exist differences, wherein the liver spectrum of protoporphyrin IX fluorescence peak was significantly higher than the abdomen spectra, and the first and second principal component of liver spectra with abdominal spectra separate accurate Third, the four main components of the upper and lower abdomen spectral classification accuracy rate of 81.82% was 100%, we can see different abdominal tissue autofluorescence spectroscopy to distinguish between the application of laser-induced fluorescence spectroscopy, assisted surgery provides possible.
