Correlation between the Loss of FHIT Gene and HPV16/18 Infection in Cervical Carcimoma and CINⅢ

Abstract


Objective :To study the role of fragile histidine triad ( FHIT) gene in occurrence and development of cervical carcinoma and the cervical intraepithelial neoplasia gradeⅢ(CINⅢ),and to analyse the relationship between the loss of FHIT gene and human papilloma virus (HPV) infection.Methods: We chosed 50 cases of normal cervical epithelial, 20 cases of CINⅢ,43 cases of cervical squamous cell carcinoma . Then,the expression of HPV16/18 DNA were determined by tissue microarrays (TMA) and in situ hybridization. The loss status of FHIT gene were examined by polymerase chain reaction (PCR) technique. To take Statistical analysis by SPSS 13.0.Results: 1. The positive expression rates of HPV16/18DNA was 6%, 55% and 76.7% respectively in normal epithelial, CIN III and cervical carcinoma. Statistical analysis showed the positive expression rates of HPV in normal epithelial significantly lower than that in CINⅢor carcinoma groups(P<0.001).And there were obvious difference between three of them (x2=49.425, P<=0.001,<0.05), as well as linear-by-linear x2 =47.742, P <0.001 .But there is no significant difference (P>0.0167) between in CIN III and in cervical carcinoma case. 2. The loss rates of FHIT gene were 0%, 45% and 72.09% in normal epithelial, CIN III and cervical carcinoma respectively. Statistical analysis showed the loss rates of FHIT gene in normal epithelial significantly lower than that in CINⅢor in carcinoma groups(P <0.001). And there were obvious difference between three of them (x2=53.532, P <0.001,<0.05), as well as linear-by-linear x2 =52.478 , P <0.001 . But there is no significant difference (P>0.0167) between the loss rate of FHIT gene in CIN III and in cervical carcinoma case. 3. In three groups , there were 29 cases of FHIT gene loss in the 47 cases of HPV DNA infection (loss rate 61.7%) ,and 11 cases of FHIT gene loss in the 66 case of HPV DNA negtive express (loss rate 16.67%). Statistical analysis suggested that the loss rate of FHIT gene in the HPV positive cases was significantly higher than that in the HPV negative cases(x2 =24.347,P <0.001). But there is no significant difference (P>0.0167) in CIN III and in cervical carcinoma recpectively .Conclusion: 1.Carcinogenesis and development of cervical carcinoma are closely related the loss of FHIT gene.2. FHIT gene is related with HPV .The loss of FHIT gene in the invasive cervical squamous cell carcinoma may play a important role in the carcinogenesis and development with HPV infection.
