Abstract


Most male deer antler is growing animals an as yet immature ossification angle, as traditional Chinese medicine in China has two thousand years of history. As a special antler cartilage is rich in a natural cytokine library active factor in its growth process from the body, bone, blood, nerve growth, fibroblasts, epithelial cells simultaneously, as deer body in its most active growing point before ossification store large quantities of growth factors, which are the biochemical basis of clinical effects velvet, velvet Modern medical research shows that a wide range of pharmacological activity. But antler growth-promoting, promoting metabolism, anti-inflammatory, immunomodulatory, brain puzzle, anti-aging, blood, wound and fracture healing is now known pharmacological effects of chemical composition can not explain. Therefore, the regulation of the antler is rich in various organs and tissues grow and develop a variety of polypeptide growth factor research is very meaningful. With national attention for the gradual modernization of Chinese medicine, we chose PAP regulation of cellular immunity were studied. One, antler polypeptide extraction and purification and determination of antler slice, carefully cut edge fluff, adding 70% ethanol, 4 ℃ soak 48 hours, 16000RPM speed centrifugation and the supernatant. The low-pressure chromatography on Superdex-75 gel, eluted with 20% ethanol, 280nm UV spectrum visible in six separate protein peaks, in batches collected eluent, to give six components, namely Ⅰ labeled component , Ⅱ, Ⅲ, Ⅳ, Ⅴ, Ⅵ. Second, the PAP on the immune function of mice was observed in this study PAP blood serum of mice of different components of interleukin-2 content, splenic T lymphocyte proliferation, NK cell activity of spleen and peritoneal macrophage phagocytosis impact and to try to gauge the role of immune activity antler polypeptides. The results showed that: component Ⅲ, Ⅳ and mixed peptide groups can significantly improve the peripheral blood of mice levels of IL-2 in vivo could significantly synergistic lymphocyte proliferation stimulated by ConA and can significantly improve mouse spleen NK cell killing activity, and reached the level of significant difference (p lt; 0.01), is the most active component Ⅳ, but component fraction Ⅲ and Ⅳ with crude extracts were no significant differences between (p gt; 0.05); peritoneal macrophages through neutral red experiment shows that fraction Ⅲ, Ⅳ and crude extract can significantly improve mouse peritoneal macrophage phagocytosis (p lt; 0.01), is the most active extracts, but coarse extract fraction Ⅲ and Ⅳ with no significant difference between (p gt; 0.05). PAP at different doses on immune function in rats found a dose-and immune regulation there is a certain correlation between the increasing concentration enhancement effect, but the difference was not significant statistical results.
