Effect of Remifentanil Preconditioning and Ischemic Preconditioning on Apoptosis of Hepatocytes Induced by Ischemia-reperfusion Injury in Rats

Abstract


Objective: To investigate remifentanil preconditioning (remifentanil preconditioning, RPC), ischemic preconditioning (ischemicpreconditioning, IPC) on rat liver ischemia-reperfusion (ischemia reperfusion, IR) injury early liver cell apoptosis . Methods: 72 healthy male SD rats were randomly divided into four groups: sham operation group (S group ) ; ischemia-reperfusion group (IR ) ; remifentanil preconditioning group (R group ) ; ischemic preconditioning group (I group ) , the production of rat liver in situ warm ischemia -reperfusion model , each group were reperfusion 1h, 3h, 5h killed six rats , according to the need to deal with liver specimens. Light microscopy of liver tissue morphological changes ; TEM cell ultrastructural changes: terminal deoxynucleotidyl transferase- mediated dUTP nick end labeling (TUNEL) assay liver apoptosis and apoptotic index calculation (apoptosis index, AI); immunohistochemical assay of liver tissue Bcl-2, Bax protein expression and using image analysis system for semi-quantitative analysis . Results : IR group , R group, I set AI values ??significantly higher than those in the Sham group (P <0.05); while the R group , I set AI values ??were significantly lower than the IR group (P <0.05): R group and I AI group had no significant difference (P> 0.05). Compared with Sham group , IR group Bax protein expression was significantly increased (P all <0.05), Bcl-2 protein expression was significantly decreased (P <0.05). Compared with IR group , R group Bax protein levels were significantly decreased (P all <0.05), I set Bax protein levels did not change significantly (P> 0.05); R group and the group I Bcl-2 protein expression were significantly increased (P < 0.05 ) . Conclusion: remifentanil preconditioning and ischemic preconditioning can reduce hepatic IR early liver cell apoptosis and reduce hepatic ischemia-reperfusion injury, remifentanil preconditioning may increase the expression of Bcl-2 , Bax inhibition the expression ; ischemic preconditioning may increase the expression of Bcl-2 .
