Objective: To investigate exogenous basic fibroblast growth factor (basic fibroblast growth factor, bFGF) tendon allograft transplantation. Methods: 108 Wister rats, weighing 220 g to 270 g, male or female. The optional 36 rats were used as donor, to take sides with the health, more than 10 days refrigerated storage at -80 ℃ deep cryogenic refrigerator prepare transplantation. More than 72 rats receptors, were randomly divided into three groups (n = 24), the removal of the side with health, take frozen the tendon bridge in tendon defect. Group A (control group): tendon allograft transplantation, the to graft both sides of the stump without any reagents: B group (negative control group): tendon allograft transplantation, transplant tendons both sides of the ends of each application fibrin blocking agent (fibrin sealant, FS) 3μl; C (experimental group): tendon allograft transplantation, transplantation the tendon both sides of the stump and bFGF and FS mixture 3μl (for containing of bFGF 500ng). 4 and 8 weeks after surgery, each group were 12 rats graft gross, histological observation and biomechanical test. Results: general observation: After four weeks, eight weeks each group the degree of tendon adhesions, A, B between the two groups showed no significant difference (P> 0.05), but the group C and A, two groups of B, the differences were statistically significant (P <0.05);, after 4 weeks, 8 weeks groups the tendon healing situation, A, B between the two groups was no difference in group C compared with A, B groups. Histological observation: after four weeks, eight weeks between the two groups, A, B, there was no significant difference in the number of fibroblasts, tendon tendon beam density, group C and A, B two groups, there were significant differences. Biomechanical test: after four weeks, the maximum tensile strength of the A, B, C group were 23.21 ± 1.98,24.59 ± 2.32,33.69 ± 2.71N maximum tensile displacement were 4.52 ± 0.43,4.38 ± 0.27,3.01 ± 0.35mm, A, B between the two groups showed no significant difference (P> 0.05), group C and A, B two groups compare each index differences were statistically significant (P <0.05); postoperative 8 weeks maximum tensile strength of the A, B, C group were 40.82 ± 3.66,41.62 ± 3.49,50.34 ± 4.81N maximum tensile displacement were 2.91 ± 0.25,2.85 ± 0.36,1.83 ± 0.17mm, A, B groups between the difference was not statistically significant (P> 0.05), group C and A, B two groups, each of the indicators of the differences were statistically significant (P <0.05). Conclusions: (1) used in the graft ends exogenous bFGF can promote the speed of the healing of the graft, but aggravated the tendon adhesions; (2) Cryopreservation tendon tendon tissue to reduce antigenicity.
