Abstract


Objective: To observe serious burns SD rats organ bradykinin B1 receptor expression changes of bradykinin B1 receptor-specific inhibitor of scalded rat organ vascular permeability changes. Methods: 90 adult SD rats quality 250 ~~ 300g, were randomly divided into normal control group (N), sham group (S group), a simple scald group (B), sham burn bradykinin B1 receptor specificity inhibitor group (SI group) and burns bradykinin B1 receptor-specific inhibitor group (BI group). S group rehydration does not scald group B and BI rats caused 30% Ⅲ degree burns and rehydration, 0 hours after injury, 3 hours, 6 hours, 12 hours, 24 hours and 48 hours of lung, heart , kidney, small intestine, immunohistochemical method observation bradykinin B1 receptor expression was measured by Evan's Blue (EB) France, tissue wet and dry gravity method organs vascular permeability changes. Results: 1, normal SD rat heart, lung, kidney and small intestine had no bradykinin peptide B1 receptor expression, the serious burns SD rat heart, lung, kidney and small intestine exists in the early stage after injury (3-24 hours) bradykinin B1 receptor expression, cell type myocardium within the inner wall of the small arteries, small veins and capillary endothelial cells, capillary endothelial cells within the airway submucosa, alveolar type I epithelial cells, renal cortex and medulla gap small vascular endothelial cells, the epithelial cells of the proximal tubule, small intestinal layer, submucosa and serosa of small vascular endothelial cells mainly give specific inhibitor des Arg9-Leu8-BK receptor expression, no significant changes. 2, severe burns SD rat heart, lung, kidney and small intestine vascular permeability after severe trauma, significant increase in basic fluid oozing peak reached within 3-6 hours after injury, and sustainable to injury after 24 hours , tissue edema sustainable throughout the shock stage, vascular permeability changes in the trend of bradykinin B1 receptor expression basically. Bradykinin peptide B1 receptor specific inhibitor des Arg9-Leu8-BK can significantly reduce the the severe burns SD rat heart, lung and kidney vascular permeability within the first 24 hours after injury, which hurt The inhibitory effect is most obvious early (3-6 hours) significantly alleviate the shock during the major organs edema. Discussion and conclusions: 1. Studies have shown that normal SD rat heart, lung, kidney and small intestine had no slow bradykinin B1 receptor expression, three hours after severe burns, immunohistochemical method you can see the heart, lungs, kidneys and small intestine and other organs and tissues bradykinin B1 receptor expression was significantly upregulated, a specific inhibitor of des Arg9-Leu8-BK receptor expression after injury. Severe burns organs and tissues of rats bradykinin peptide B1 receptor expression distribution in heart intramyocardial the inner wall of the small arteries, small veins and capillary endothelial cells; lung airway submucosa capillary endothelial cells I alveolar epithelial cells mainly; kidney cortex and medulla gap vascular endothelial cells, epithelial cells of the proximal tubule-based; small intestine mucosa, submucosa and serosa vascular endothelial cells. 24 hours after severe burns SD rat heart, lung, kidney and small intestine vascular permeability after severe trauma, significant increase in basic fluid oozing peak reached within 3-6 hours after injury, and sustainable to injury , tissue edema sustainable throughout the shock stage, vascular permeability changes in the trend of bradykinin B1 receptor expression basically. Bradykinin peptides the B1 receptor specific inhibitors des Arg9-Leu8-BK can significantly reduce serious burns SD rat organs and tissues within the first 24 hours after injury vascular permeability, which early after injury (3 -6 hours) inhibitory effect is most obvious, edema significantly alleviate the major organs of the shock period. 5. Above results, SD rats were scald major organs slow bradykinin B1 receptor expression in a significant increase in vascular permeability kinin system by bradykinin B1 receptor-mediated the serious burns SD rats each exudation increased occurrence of the main organs of the organization, the use of specific inhibitors of the receptor can effectively inhibit and reduce the occurrence of exudation and edema of the tissues and organs of the body fluids. These results for the search for effective prevention and treatment of severe burn shock and organ edema drugs provide a new target.
