Objective: To investigate the prostaglandin E1 (PGE1) on endotoxin lipopolysaccharide (LPS) -induced rat Kupffer cells (Kupffer Cell, KC) secretion of inflammatory cytokines affect . Of rat Kupffer cells stimulated with LPS and PGE1 role under changes in the level of inflammatory cytokines and nuclear factor -kB ( NF -kB ) , to further explore the anti-inflammatory effect and mechanism of PGE1 , provide new inspiration for clinical application . Methods: collagenase cyclic perfusion and Nycodez of density gradient centrifugation method to obtain high purity rat Kupffer cells, washed twice After overnight incubation , the cell concentration of 4-6x105/ml , seeded in 24 - well culture plate in situ . Isolated and cultured Kupffer cells 48 hours , respectively, after the establishment of different concentrations of PGE1 group , to establish a non-toxic dose range of cells . Added to the Kupffer cells of LPS and PGE ( three different concentrations) , divided into a control group , stimulation group (LPS 10ng/ml) and PGE1 group (LPS PGE1) stimulation of primary cultured Kupffer cells three hours . The effects of different concentrations PGE1 rat Kupffer cells , TNF-α ( tumor necrosis factor -α) and IL-6 ( interleukin-6 ) release . LPS and PGE1 stimulation and observe the activity of NF-kB in Kupffer cells . The enzyme-linked immunosorbent assay (ELISA) detection of TNF-α, IL-6 level was measured by radioimmunoassay (RIA) . Detected NF-kB activity and specificity EMSA gel electrophoretic mobility shift assay (EMSA) . Results: The normal rat Kupffer cells in vitro after LPS stimulation , NF-kB activity significantly enhanced the supernatant of TNF-α and IL-6 secretion were significantly higher ( P lt; 0.01 ) ; different the concentration group PGE1 can significantly reduce TNF -α , IL - 6 release level and activity of NF-kB (P lt; 0.05), and the dose-effect relationship significantly . Conclusion: The acute and chronic liver injury caused by various diseases , LPS through the activation of NF -kB , further activate TNF -α , IL -6 expression of inflammatory cytokines . Excessive activation of Kupffer cells , the release of a large number of inflammatory cytokines , high levels of endotoxin blood lead endotoxemia , etc. , plays a very crucial role in the pathogenesis . PGE1 can by down-regulating the activity of NF-kB , and reduce the excessive activation of Kupffer cells , reduce TNF -α expression of IL -6 and other inflammatory cytokines , reduce the high levels of endotoxin in the blood and other aspects of regulation of the body play a protective role .
