Abstract


Stromal cell derived factor (stromal cell derived factor-1α, SDF-1α) as a chemokine to its receptor CXR4 in embryonic development and metastasis of tumor cells and play an important role mediated immunodeficiency virus (HIV) infection . Recent studies have found that SDF - 1alpha is highly expressed in atherosclerotic plaques , while its closely related to the vessel wall inflammation . In visible SDF-1α/CXCR4 in atherosclerosis atherosclerosis occurs role in the development process can not be overlooked . This experiment explores SDF-1α/CXCR4 on monocyte chemotaxis , monocyte and endothelial cell adhesion and advanced oxidation protein products (Advanced oxidation protein products, AOPP) the role ; Second, the study AOPP endothelial cells SDF - 1alpha expression . Objective: To investigate SDF - 1alpha , monocyte chemoattractant monocyte chemotaxis and AOPP ; SDF-1α/CXCR4 the ECV304/THP-1 adhesion ; AOPP SDF - 1 alpha expression in endothelial cells . Method : small chamber migration experiments to study the SDF - 1alpha role chemoattractant for monocytes , mononuclear cells or by AOPP processing a single nuclear cell to join the upper chamber , the lower chamber were incubated for 10 hours Add SDF - 1alpha count each group chamber the number of cells migrated . The adhesion experiment to study the adhesion of THP-1/ECV304 ECV304 cells with 50μmol / l AOPP incubated for 24 hrs, the THP-1 cells were incubated with ECV304 cells eluted after detecting adhesion . RT-PCR analysis of SDF - 1alpha mRNA expression . AOPP observed a dose-dependent effect of SDF - 1 alpha expression on ECV304 cells . Results : 1.SDF-1α concentration-dependent induction of monocyte migration , CXCR4 antibody significantly inhibited this effect ; the AOPP 24 hours after treatment with different concentrations of THP-1 cells and then the SDF - 1 alpha chemoattractant and found AOPP a dose - dependent increase in the migration of THP-1 cells . The 2.AOPP a dose- dependent increase adhesion of THP-1/ECV304 . 3 in the range of 0-200μmol / L , AOPP was concentration-dependent upregulation of SDF - 1 alpha expression . Conclusion : 1.SDF-1α/CXCR4 involved in the chemotactic migration of mononuclear cells , whose role is AOPP strengthened. 2.SDF-1α/CXCR4 involved in monocyte / endothelial cell adhesion AOPP strengthen its role . 3.AOPP can upregulate endothelial cell expression of SDF - 1alpha .
