Abstract


Objective: To investigate the D - limonene (d-limonene) in human leukemia cell apoptosis effects and mechanism . Methods: A human leukemia cell line K562 and HL-60 cells for the study using thiazolyl blue (MTT) and trypan blue staining to detect the impact of D - limonene on cell proliferation and Annexin V / PI double staining of cells withered the impact of death associated protein expression by western blot assay mitochondrial apoptotic pathway . Results : d- Limonene role K562, HL - 60 cell apoptosis rate were time and dose-related increase (0.4 mmol / L after 72 hours up to 71.21% ); treated Bcl-2/Bax ratio decreased ( down from 1.93 to 0.75 ) , increased cytoplasmic cytochrome c expression significantly increased the expression of caspase -9 and caspase-3 split . Conclusion : d-limonene can induce apoptosis of leukemia cells ; activation of the mitochondrial apoptotic pathway might be its main mechanism of apoptosis induced .
