Abstract


Purpose of this experiment is the use of two kidney one clip renal hypertensive rats , observed reactive oxygen thoracic aortic reconstruction , as well as the application of Tempol ( 4 - hydroxy - 2.2.6.6 - tetramethyl- piperidine ) after intervention effect , further explore Tempol impact on the function and structure of the renal hypertensive rat aortic and mechanism of action . Established method two kidney one clip renal hypertensive rats after 4 weeks were randomly divided into sham group (n = 8), and hypertension group ( n = 6 ) and treatment group ( n = 6 ) . The treatment group received containing Tempol 1 mmol / L drinking water , observed 8 weeks after the administration of blood pressure, angiotensin Ⅱ (Ang Ⅱ), nitric oxide ( NO ) , 8 - iso - prostaglandin F2α ( 8 -iso -PGF2α ) , chest aortic NADPH oxidase subunit p22phox mRNA expression changes ; diastolic function and structure of isolated vascular ring experiments and HE staining observed changes in the thoracic aorta . 1 hypertension group compared with the sham group : a significant increase in blood pressure , Ang II , 8 -iso - PGF2α film thickness in the aortic media thickness / inner diameter ( P lt; 0.01 ) ; of NO significantly reduce ( P lt ; 0.01 ) ; isolated aortic rings induced maximal relaxation to acetylcholine (Ach) percentage decreased significantly (P lt; 0.01); aortic p22phox mRNA upregulation . 2 , after eight weeks of treatment in the treatment group with Tempol compared with the hypertension group : drop in blood pressure ; the aortic membrane thickness , media thickness / internal diameter decreased ; 8-iso-PGF2α significantly decreased ( P lt; 0.01 ) ; the NO level rise ( P lt; 0.05); percentage of maximal relaxation of isolated aortic rings Ach caused a significant increase ( P lt; 0.01 ) ; aortic p22phox mRNA downregulation ; Ang Ⅱ relatively no difference . 3 , isolated aortic rings in each group the maximum diastolic percentage of sodium nitroprusside caused no difference , with L- nitro - arginine methyl ester (L-NAME) to inhibit NO production , maximum relaxation response to Ach no difference between . Conclusion renal hypertensive rats of Ang Ⅱ , NO and oxidative stress involved in the hypertensive aortic function and changes in the structure ; can significantly reduce the level of oxidative stress in renal hypertensive rats Tempol improve NO metabolism , can lower blood pressure its antihypertensive mechanism related to the improvement of NO metabolism are closely related , Tempol also improve aortic endothelium-dependent relaxation down of aortic p22phoxmRNA expression .
