Abstract


Purpose: AMP579 a new synthetic adenosine analogues, has a role, long half-life, and other features. Adenosine on coronary heart disease in patients with inflammatory cytokines such as tumor necrosis factor (TNF-α), interleukin -6 (IL-6) and interleukin -18 (IL-18) studies have been reported, but no AMP579 leukocyte differentiation antigen 40 ligand (CD40L) role in the study reported. Recent studies have found that the CD40L ligand as important inflammatory signaling pathways play an important role in atherosclerosis development. The purpose of this project is to a more comprehensive study the AMP579 of the peripheral blood of patients with acute coronary syndrome (ACS) soluble leukocyte differentiation antigen 40 ligand (sCD40L) expression, compared with the same concentration of adenosine's role, to explore different concentrations AMP579 and adenosine on the role of differences in the peripheral blood of patients with ACS sCD40L expression and its mechanisms; AMP579 treatment of coronary heart disease in clinical practice to provide a theoretical basis and determine the appropriate therapeutic dose for clinical guidance. Methods: The clinical symptoms, ECG changes and serum creatine kinase diagnosed 15 patients with acute myocardial infarction (AMI) patients, 15 patients with unstable angina (UA) patients and 15 normal subjects, using enzyme-linked immunosorbent assay reaction (ELISA) sCD40L expression in the peripheral blood of patients with quantitative detection, while using different concentrations of AMP579 and adenosine peripheral blood intervention study of sCD40L expression in the peripheral blood of patients, and sCD40L correlation analysis of risk factors for coronary heart disease . Results: ① normal peripheral blood of patients with UA and AMI sCD40L was significantly increased, a significant difference (F 451.066, p lt; 0.05), AMI of sCD40L expression level in patients than UA patients, a significant difference (p lt; 0.05). ② UA group, AMP579 and adenosine on peripheral blood the sCD40L express role overall significant difference (F 49.610, p lt; 0.01); AMI group, AMP579 and adenosine expression on peripheral blood sCD40L overall significant The difference (F 171.362, p lt; 0.001); in normal group, AMP579 and adenosine on peripheral blood sCD40L expression of the role of non-significant difference (F 0.584, p = 0.446 gt; 0.05). Semi-logarithmic chart analysis shows: in the UA group, AMP579 and adenosine on peripheral blood the sCD40L express role were reduced with the increase of the concentration of the drug, compared with adenosine, AMP579 decreasing trend is more obvious; AMI group, AMP579 sCD40L expression of the role of adenosine on peripheral blood increased as the concentration of the drug decreased compared with adenosine, the AMP579 reduce the trend is more obvious. ③ give different concentrations of the peripheral blood of patients with AMI AMP579, concentrations were 0 μmol / L, 110 μmol / L, and 330 μmol / L, 1.1 × 103μmol / L, and 3.3 × 103μmol / L, and the resultant the sCD40L value of in general have a significant difference ( F is 501.059 p LT; 0.001), and there was significant difference in the concentration between any two; AMP579 give different concentrations of the peripheral blood of patients with UA, the concentrations were 0 μmol / L, 110 μmol / L, 330 μmol / L, 1.1 x 103μmol / L, 3.3 × 103μmol / L AMP579, the sCD40L value obtained in general there is a significant difference (F 919.114, p LT; 0.001). There was no statistical difference in the concentration of 1.1 × 103μmol / L and 3.3 × 103μmol / L, there were significant differences between any two of the remaining concentration. Conclusion: ① ACS patients sCD40L expression than normal was significantly higher, and the peripheral blood of patients with AMI sCD40L expression level higher than UA patients, suggesting that sCD40L can be used as an important indicator of the ACS condition the judge and prognosis estimated.
