Objective To study the spinal cord antinociceptive effect in the rat model of formalin-induced pain Intrathecal midazolam median effective dose (ED 50 ) and c-fos expression in the dorsal horn of the spinal cord impact to explore midazolam dose analgesia in formalin-induced pain rat model of spinal cord level, to provide new ideas for spinal administration. Methods The experiment is divided into two parts (1) sequential determination of intrathecal injection of midazolam on formalin-induced pain in rats spinal analgesia ED 50 values. (2) intrathecal injection of midazolam on formalin-induced pain rat spinal cord c-fos expression. Select intrathecal catheter successfully 32 male SD rats were randomly divided into four groups (n = 8): control group (C group), saline group (NS), before the pain caused by the administration group (M 1 group) and cause pain after administration group (M 2 ). Results (1) stability in the formalin-induced pain in a rat model of intrathecal injection of imidazole semi-effective when spinal cord antinociception of effective dose (ED 50 ) 21.8μg; (2) the rats in each group flinching, licking cumulative time comparison, NS group, M , 1 the group M 2 group flinching, licking cumulative time was significantly longer in group C (P <0.01); the the M 1 the group M 2 group flinching, licking the accumulated time is shorter than the NS group (P <0.01); and M 1 group is shorter than the M 2 group (P <0.01); (3) Group C, the NS group spinal cord dorsal horn c-fos expression was increased (P <0.01 c-fos expression dorsal horn of the spinal cord); compared with NS group, M 1 group M 2 group lower (P <0.01); M 1 the group M 2 Group spinal cord dorsal horn c-fos expression was increased (P <0.01). Conclusion (1) in formalin-induced pain in a rat model of intrathecal injection of midazolam in the spinal cord antinociceptive effect half way effective dose 21.8μg, the analgesic effect is more precise. (2) midazolam preemptive analgesia effect.
