Stevia (steviol glycosides) are a class from the Compositae stevia (Stevia rebaudiana Bertoni) leaves extract obtained high sweetness and low calorie natural non-nutritive sweeteners. Stevioside has a variety of pharmacological activity in vivo without toxic side effects have been used as a natural sugar substitute, has aroused the attention of scholars. Currently, stevia is widely used in cosmetics, food and pharmaceutical industries. In this thesis, stevioside as a research object, the extraction rate of stevioside, stevia loss rate and purity stevia evaluation around stevia extract, purify, purification, refining and bleaching process conditions optimized for the study Target carried out the following aspects of work: 1. stevia extraction technology research evaluation stevioside extraction rate were investigated reflux extraction, extraction method, countercurrent extraction, enzymatic and reflux formulation five kinds of sets method to extract stevia leaves stevia extraction process. The results showed that: reflow method, extraction method, enzymatic, reflux and reflux method sets the reference to the optimum extraction conditions, the extraction rate and 94.54%, respectively 98.08,80.17,86.98,79.98. Compared reflux sets formulation of extraction rate, time is short, less water consumption. Therefore, the return set formulation is used to extract stevia has unique advantages. 2 stevia glycosides stevioside Purifying Technology study the loss rate and the impurity yield index, investigated several methods purify water extract of stevia impurity effect, the experimental results show that the alcohol precipitation method Sweet chrysanthemum leaf water extract of impurity with good results, in addition to alcohol and alcohol precipitation Shen optimum conditions were: 10 mL / min speed to the water extract of 95% ethanol was added to the system, up to 80% ethanol concentration, static Set 1 h, centrifuged. Under these conditions, the optimum rate of 16.47% impurity, sweet glycosides loss rate of 6.13%. 3 stevioside Purification Process of (a) through dynamic adsorption experiment, the system of L macroporous resin stevioside best adsorption / desorption process conditions: using a series double column, sample volume of 4200mL ; the sample density 0.025g/ml; adsorption rate of 40 BV / h; eluted with water an amount of 800 mL (10BV), elution rate of 7.2 BV / h; adsorption and elution temperature was 40 ℃; eluent mixed solution; elution rate of 20 BV / h. (2) Select the four kinds of macroporous resin, stevioside by comparing its results of static adsorption screened more suitable for refined stevia macroporous resin, and for the dynamic adsorption of stevia, the process conditions: on concentration of 0.1 g / mL; adsorption rate of 12 BV / h; the sample volume 40 mL; 70% ethanol as eluent, collecting 100 mL. 4 stevioside bleaching technology of active alumina - medium pressure column adsorption decolorization on stevia, stevioside found on activated alumina has some bleaching effect on the purity and quality of stevia also have some impact. The optimum conditions are as follows: water as solvent; adsorption temperature of 35 ℃, adsorption time of 30 min; stevioside alumina mass ratio of 1: 0.8; stevioside after bleaching medium pressure column chromatography using a continuous method Stevia glucosidase purified preparation, elution with distilled water, and the resulting purity of stevioside 91.76%.
