Abstract


Riboflavin is one of the 13 vitamins that are necessary for human health. It is engaged in matter and energy metabolism as an important composition of prosthetic group of many enzyme system. Shortage of riboflavin may cause important effects on human body. There are four main sythetic method for riboflavin including chemical synthesis using D-glucose as raw material, semisynthesis, saccharomycete fermentation method and genetic engineering fermentation method. Nowerdays, semisynthesis, saccharomycete fermentation method and genetic engineering fermentation method are mainly adopted for industrial production.This paper studies on the process of semisynthesis through 4 sections and makes suggestion on process improvement.(1) Change the solvent used for conjugates content test. Traditional conjugates content test uses formic acid as solvent which has the disadvantages of strong irritation and corrosivity and relatively high price. Methonal is used as solvent instead of formic acid in this paper. The solubility of conjugates in methanol satisfies the test. And the conjugates with methanol.is scanned in ultraviolet The result shows that match-mismatch absorption peaks appear in 325.50nm In this condition, absorbing degrees of conjugates with different methanol concentration are test.Linear equations is obtained, Y=0.01+47.15X, R=0.9993. The result shows that using methonal as solvent is feasible.(2) Determine the optimum consumption of glacial acetic acid during cyclization process. Glacial acetic acid used in cyclization experiment is a mixture of recycled butyl acetate containing part of acetic acid and fresh glacial acetic acid as catalyst. This paper studies on different ratio of butyl acetate and acetic acid to optimize the consumption of glacial acetic acid. The experiments prove that 70% of butyl acetate and 10ml glacial acetic acid as appropriate dosage.(3) Optimize the reaction time and temperature of cyclization reaction. In present process, the cyclization reaction lasts for 9 hours under temperature of 115℃. By changing the reaction time and temperature, this paper obtains conclusion that integratively consider from economy aspects, the optimized condition for cyclization is with reaction time of 7 hours under temperature of 115℃.
