Abstract

Oxya chinensis(Thunberg)(Orthoptera:Oedipodidae) is a common species of Oxya Serville and is one of the most major agricultural pests. It feeds on rice, wheat, corn, grass, broomcorn, bulrush and any other gramineous plants. In our dissertation,we tested and compared the activity of detoxification enzyme and target enzyme of Oxya chinensis fed on rice, wheat, grass respectly, meanwhile,we compared the susceptibilities to malathion of Oxya chinensis fed on different host plants to discuss the relationship of plants, pests and insectcides.(1) Comparisons of the enzymes activity of the fourth-instar nymph of Oxya chinensis fed on different host plantsWe studied the specific activity of esterase(EST),glutathione S-transferase(GST),acetylcholinesterse(ACHE) in the fourth-instar nymph of Oxya chinensis fed on three host plants(grass(Setaria viridis),wheat (Triticum aestivum),rice(Oryza sativa)) for different days(1d,3d,5d).The results showed that the activity of EST,GST,ACHE were affected by the host plants obviously. For EST,there were significant differences(P<0.05) among Oxya chinensis fed on three host plants and that fed on rice had the highest EST activity. For GST,Oxya chinensis fed on rice was significantly higher than that fed on wheat for 1d and 3d.Oxya chinensis fed on grass had the highest GST activity for 5d,this may have the relationship with the special physiological state that Oxya chinensis might transform into the next development stage after fed on different plants for 5d.For ACHE,ACHE activity of Oxya chinensis fed on rice was significantly higher than that fed on wheat and grass. For the same plant, there were no significant differences among the EST activity of Oxya chinensis fed on for different days, and the EST activity of Oxya chinensis fed on different host plants had different hydrolyzing ability for different substrates. The GST activity of Oxya chinensis fed on the same plant for different days depended on the gender of Oxya chinensis and the kinds of the plants fed on.The AChE activity of Oxya chinensis fed on the same plant for 1d was significantly higher than that for 3d and 5d.(2) Effects of Host Plants on the Activity of Enzymes of Oxva chinensis and its sensitivity to insectcidesThe results showed that the sensitivity sequence of Oxya chinensis fed on different host plants to malathion was grass>wheat>rice, the activity of EST and GST of Oxya chinensis fed on rice were the highest among the three host plants. Therefore, the differences of the activity of EST and GST of Oxya chinensis fed on different host plants was one of the reasons related to its different sensitivity to malathion. The AChE activity of Oxya chinensis fed on rice was the highest among the three plants in both female and male, the In vitro inhibition of Malaoxon of AChE of Oxya chinensis fed on different host plants also showed that the sensitivity sequence was wheat>grass.>rice.So it may be inferred that different allelochemicals in different plants have various affection on AChE activity of Oxya chinensis, and this affection resulted the different sensitivity of Oxya chinensis to insecticides.(3) Effects of Host Plants on the Activity of Enzymes of Oxya chinensis in different development stageWe studied the specific activity of EST,GST,AChE of Oxya chinensis fed on different host plants(grass, wheat, jindao 8,jindao 9,handao297) from the first-instar nymph to the fifth-instar nymph. The results showed that the effects of different host plants on the detoxification enzyme and the target enzyme of Oxya chinensis in different development stage was depended on the gender and the development stage of Oxya chinensis and the kinds of the plants fed on.
