Abstract


Apoptosis is the main form of cerebral hemorrhage surrounding tissue cell death. The mitochondrial apoptosis Center. Changes in mitochondrial ultrastructure and above the two are closely related. So explore ICH animal models around the hematoma neurons, mitochondrial ultrastructure the pathological changes of great significance. On this basis, to explore the internal mechanism of rhubarb treatment of acute cerebral hemorrhage. Objective: To observe the ICH hematoma surrounding neurons dynamic changes of mitochondrial ultrastructure explore rhubarb treatment of ICH possible mechanism of action. Method: adult Sprague-Dawley rats were injected in the caudate nucleus collagenase Ⅳ (0.05U/0.5uL) induced ICH, sham-operated rats saline injection of the same dose in the same position. After 3h, 6h, 12h, 24h, 48h, rats excessive anesthesia, cardiac perfusion brain. Randomly photographed using electron microscopy, qualitative observation of the neuronal mitochondria perihematoma ultrastructure change. In situ end (TUNEL) method labeled apoptotic cells around the hematoma. Intracerebral hemorrhage in rats ICH 3h, 6h, 12h, intraperitoneal injection of rhubarb Injection (5ml/kg) of saline or the same dose. Rat brain hemorrhage after 3h and 6h give rhubarb injection significantly reduce mitochondrial ultrastructural lesions inhibit casepsle-3 activation (P <0.001), and reduce the TUNEL-positive cells (P <0.05), and reduction of apomorphine the number of rotations (P <0.05); induced cerebral hemorrhage after 12h administration is invalid. Conclusion: cerebral hemorrhage early visible pathological changes of mitochondrial ultrastructure; 2. Perihematoma apoptosis rate peaked at 24h after cerebral hemorrhage; 3. Rhubarb may play a neuroprotective by blocking the mitochondrial-dependent apoptosis pathway role.
