Abstract

This thesis include the ginsenosides Rb 3 the microbial transformation intends of Penicillium sp.229 metabolites of two parts. After a long research, the lab is isolated from the soil of ginseng growing areas screened Paecilomyces contains a lot of glycosidase mutagenesis screening proposed Penicillium sp.229, can be efficiently converted ginsenoside Rb 1 rare ginsenoside Compound-K (CK), the mole conversion rate above 75%. Ginsenoside Rb 3 belong to the same glycols ginsenosides, Rb 1 structure is very similar in nature, especially in higher levels in the stem and leaf, but the current there was no reported microbial transformation of ginsenoside Rb 3 . This study explored ginsenoside Rb 3 HPLC separation method, and the establishment of the standard curve, stem and leaf saponins Rb 3 content determination Determination of stem and leaf saponins Rb 3 content. Saponins while the stem and leaf by column chromatography separation, after a normal phase silica gel and the inverted repeated chromatography on silica gel was prepared to give the purity of more than 95% of the ginsenoside Rb 3 As Next microbial conversion substrate. Accumulate a sufficient amount of ginsenosides Rb 3 substrates the ginsenosides Rb 3 preparation of the amount of Fermentation and fermentation products were separated and identified, explore ginsenosides Rb 3 metabolic pathways. Metabolites have been identified of ginsenosides Mx, and ginsenoside CK, which for the first time ginsenosides Mx X-ray diffraction analysis, and the establishment of the ginsenosides Mx HPLC separation conditions and standard curve. Since then, further study of ginsenoside Rb 3 metabolic pathways, respectively fermentation product of a different time, different time crude enzyme transformation products directly immobilized enzyme conversion product of different times, different inputs substrate fixed The enzyme conversion product by TLC and HPLC analysis, analysis the ginsenosides Rb 3 metabolic characteristics, exploring the exchage. Inferred ginsenoside Rb 3 in the role Paecilomyces sp.229 may exchage: Rb 3 → Gypenoside IX → Mx → CK. Fermentation conditions to optimize the research continues ginsenosides Rb 3 , respectively, carbon, nitrogen, screening, and explore the growth factor, the impact of the surfactant on the fermentation by orthogonal method ginseng saponin Rb 3 six factors of the fermentation conditions were optimized to determine the best conditions for the conversion: 3% sucrose, 3% yeast extract, 0.1% wheat bran powder, 0.1% MgSO 4 , 0.15? Cl 2 and 0.2% (NH 4 ), 2 SO 4 , g molecular conversion efficiency up to 28.2%. Microbial metabolites due to its rich, more and more attention of researchers, microbes also produce many other active compounds, including the compound of the structure of the flavonoids, alkaloids, saponins, etc. In addition to antibiotics, has been found. The microbial metabolite has become one of the important sources of drug treatment of cancer, AIDS, cardiovascular and cerebrovascular disease. This study Paecilomyces sp.229 metabolites preliminary separation, two steroidal compounds isolated after repeated column chromatography, and preparation was obtained as crystals, X-ray diffraction analysis, was identified as a peroxygen ergost and ergosterol. This is the first time from the strains isolated two compounds, and provides a new way for their preparation.
